| Objective:(1)To study differential expression of miRNA-130a in the obesity and non-obesity people of xinjiang uygur population,and make correlation analysis.(2)To clearfy the function of mir-130a regulate the adipocytes differentiation in the 3T3-L1 adipocytes,and to anylize the relationship of mir-130a with the gene of differentiation.Methods:(1)5 obesity and 5 non-obesity,age and sex matched non-obesity from Uygur population were enrolled,differential expression of mir-130a of peripheral anticoagulant blood were performed with microarray analysis,then 21 of anticoagulant blood samples with obesity,17 samples with non-obesity from Uygur population were enrolled.Data on height,body weight,waist circumference(WC)and hip circumference(HC)were collected.Fasting plasma glucose(FPG),triglyceride(TG),total cholesterol(TC),high density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterol(LDL-C)were determined.And further mir-130a confirmed by quantitative real-time RT-PCR.(2)Using the traditional "Cocktail" induced 3T3-L1 preadipocytes to differentiate into adipocytes,using Real-Time PCR to detect mir-130a expression.Subsepuently,over-expressing mir-130a in the adipocyte,westernboting detect the expression of PPARy protein,the expression of adipogenesis marker LPL、Glut4、PPARy、C/EBP-awere analysed by Real-time PCR.Results:(1)The expression of miRNA130a in anticoagulant blood with obesity group decrease compared to non-obesity group in Uygur population.(1.14±0.32 vs 0.28±0.13,P<0.05)in microarray analysis.The data with body-mass index(BMI),WC,HC and systolic blood pressure(SBP)in Uygur obesity group was higher than nonobesity group(P<0.05).(3)The level of miRNA130a expression in obesity group was lower than non-obesity group(3.43±0.88vs1.23±0.35,P<0.05)in real-time RT-PCR.Partial correlation analysis showed that the level of miRNA130a expression in anticoagulant blood in Uygur population was negative correlated with FPG and CHOL,was positively correlated with HDL-C.(4)Cell levehto compare with 3T3-L1 preadipocytes,The level of miRNA130a expression in adipocytes has signifieantly reduced(,P<0.05).the protein of PPARγ has significantly reduce(5).After over-expression of mir-130a in 3T3-L1 preadipocytes,the expression levels of adipocyte marker genes LPL、Glut4、PPARγ、C/EBP-α and the number of lipid droplets had obviously declined,eompared with that in concrol.Conclusion The expression of mir-130a in peripheral anticoagulant blood of Uygur obesity population has significantly reduced,the expression of mir-130a may be related to Uygur BMI.(2)mir-130a suppresses adipogenesis maybe through inhibiting the expression of PPARγ. |
