In this paper,the antioxidant activity,preparation and analysis method of Salvia miltiorrhiza compound polysaccharide were systematic studied.The antioxidant activity of Pinus koraiensis pine cone pigment from waste resource were studied,the main results of the study are as follows:1.To study the antioxidant activities of Salvia miltiorrhiza compound polysaccharide and explore preliminary the relationship between polysaccharide by removing the protein and pigment and its activity.In this experiment the Salvia miltiorrhiza compound polysaccharide were prepared by using water-extraction and alcohol-precipitation method and purified further by removing the protein and pigment.The antioxidant activities of all polysaccharide products were investigated using various assays in vitro,including hydroxyl radicals,DPPH radicals scavenging ability and reducing power.Results showed that Salvia miltiorrhiza compound polysaccharide exhibited antioxidant activity in vitro,with its concentration increasing the activity of polysaccharide enhanced.The antioxidant activity of total polysaccharides became stronger after purifing by deproteinization and decoloration.The purification of Salvia miltiorrhiza compound polysaccharide still had clear concentration-response relationship.Body lipid peroxidation is one of the common causes of many diseases,and the antioxidant activity of natural ingredients often has some correlation with antitumor activity,and associates with many diseases prevention and control.The antioxidant activity of Salvia miltiorrhiza compound polysaccharide is worth further researching and utilizing.2.Establish a new method for the determination of the Salvia miltiorrhiza compound polysaccharide granules-amylase method combined with DNS method.It overcame the interference of starch to the determination of the effective component polysaccharide and the literature has not been reported in similar work.When determining the content of the Salvia miltiorrhiza compound polysaccharide granules by DNS method,we encounter difficulties.Because the starch is polysaccharide and the results of the DNS method is real reaction of the total polysaccharide content.The total polysaccharide was include the effective components of polysaccharides and starch.There was no corresponding solutions in the literature.In this experiment,we try the following ideas:First remove the starch of the granules,then converts the starch into small molecule disaccharides with the amylase method.To use the alcohol soluble difference between the polasaccharides and disaccharides,the polysaccharide was separated by alcohol precipitation method.To determine the content of the Codonopsis pilosula compound granules by the improved DNS method.It can avoid the interference of excipient starch.The results show that combined amylase method and DNS method for determination of polysaccharide in Salvia miltiorrhiza compound polysaccharide granules can overcome the interference of starch to the determination of the effective component polysaccharide.The result of methodology determination met the requirements.This method can be used for the determination of polysaccharides in Salvia miltiorrhiza compound polysaccharide granules and provide reference for other polysaccharide determination in chinese medicine preparation with starch as excipient.3.On the basis of active researching,preparation technology and quality inspection of Salvia miltiorrhiza compound polysaccharide granules were further studied.In the forming process,the hygroscopicity,fluidity,formability and other index were systematic investigated by the contrast test.Optimization the proportion of the granuals' excipient species and raw material.With hydrous ethanol as the wetting agent for wet legal system grain,drying,granuletion,Salvia miltiorrhiza compound polysaccharide granules were obtained.The experimental results can provide a reference for the research and development of salvia miltiorrhiza compound polysaccharide preparation.4.In addition to the above series researching of Salvia miltiorrhiza compound polysaccharide,the antioxidant activities of Pinus koraiensis pine cone pigment prepared by different methods were determined.The antioxidant activities of all pigment products were investigated using various assays in vitro,including hydroxyl radicals,superoxide radicals scavenging ability and reducing power.The experimental results showed that the coarse pigment extract and purified by ion exchange resin of NaCl content had obvious antioxidant activity in vitro.The antioxidant activity of NaCl eluting content were extracted by pH gradient were determined,of which 5%NaHCO3 and 5%Na2CO3 extract active components were stronger,5%NaOH extract active components below the two extracts.Known 5%NaHCO3,5%Na2CO3 extract acid were higher than 5%NaOH extract acid,thus the experiment strong acidic component in antioxidant activity were stronger.Different elution ion exchange resin to purify the hydrochloric acid-ethanol content of the experimental results showed that after the number 3 and number 2 and elution of active components,the elution of elution number 1 activity is lower than the above two components.From the above knowable,the antioxidant activities of Salvia miltiorrhiza compound polysaccharide were studied,finding out that the antioxidant activity of polysaccharides purified by deproteinization and decoloration became stronger.Studied the preparation of Salvia miltiorrhiza compound polysaccharide granules,and successfully established a new method for the determination of the Salvia miltiorrhiza compound polysaccharide granules.It overcame the interference of starch to the determination of the effective component polysaccharide,providing series of reference data for the activity,preparation and analysis method of Salvia miltiorrhiza compound polysaccharide.In addition,the antioxidant activity and primary structure of Pinus koraiensis pine cone pigment from waste resource were preliminary studied.The above work has not been reported,so I hope to provide reference for the similar research.
