Traditional Chinese medicine,plays the role of a bridge to connect traditional medical science and clinical practice.It is inevitable to discard the dross and select the essential to acquire its bioactive fundamentals.Which industrialized technology to adopt and how to evaluate the separation impact reasonably is the current problems we are facing.Macroporous resin absorption is a technique that make use of organic polymer sorbent to adsorb active ingredients from solution.And it has displayed unique effect in extraction separation of Chinese medicinal herb compared to other separation method.Besides,to adopt macroporous resins combination to break the limit of selective adsorption with single macroporous resin and achieve multi-composition optimalizing separation.It has great significance for questing the suitability of modern separation techniques to complex system of systems and realize equivalent separation of traditional Chinese medicine.In this paper,we select Hunaglianjiedu Tang as a subject investigated,for its material base has been researched deeply and with specific pharmacological action tocerebral ischemia.Based on the earlier research,we use macroporous resins combination to refined Hunaglianjiedu Tang and gain its anti-cerebral ischemia active fraction.Under the guidance of the theory of "correspondence between syndrome and prescription",we research pharmacokinetics and pharmacodynamics about the active fraction.By means of structure integrated PK-PD modeling,the relativity of pharmacokinetics and pharmacodynamics had been evaluated.It is in order to lay the foundation.of rational use of drugs and make improvement of dosage forms in clinical application about Hunaglianjiedu Tang.Objective:To explore refined conditions of macroporous resins combination to HunaglianjieduTang,to achieve desirable separation method of Chinese herbal compound;Based on the theory of "correspondence between syndrome and prescription",to research the differences of pharmacokinetics and pharmacodynamics about the active fraction in cerebral ischemia model and normal rats.To supply reference for rational use of medicines and clinical application about Hunaglianjiedu Tang.Substance:1)Select macroporous resins and their combinations to refined water extract of Hunaglianjiedu Tang,and to get each group's raffinate;by means of pharmaceutical screening of cell,to gain its anti-cerebral ischemia active fraction.2)Research of pharmacokinetics of primary ingredients such as berberine,palmatine,baicalin and geniposide in Hunaglianjiedu Tang active fraction and pharmacodynamics of anti-free radicals in cerebral ischemia model and normal rats.Method:1)Adopt 11 kinds of macroporous resin(such as X-5)with different polarity and 6 kinds of macroporous resins combination to refined water extract of Hunaglianjiedu Tang,detect contents of index constituents in different groups before and after purification by HPLC.Get adsorption-elution rates and curve of the adsorption-elution reach by calculation.At the angle of divert of chemical composition,compare the refined effect of each group.Carry out pharmaceutical screening of cell,compare the protect effect to oxygen-glucose deprivation PC12 cell of each group.Through analysis by synthesis,we may gain Hunaglianjiedu Tang anti-cerebral ischemia active fraction.2)Male SD rats use electricity to make cerebral ischemia model and with normal rats were intragastric administration same dose drug.After oral of same medicine gather the blood to heparin sodium anticoagulants centrifugal tube in stipulated time.To establish the method to determine the content of berberine,palmatine,baicalin and geniposide in plasma after pretreatment.Research pharmacodynamics of anti-free radicals simultaneously,determine content of SOD and MDA in plasma.To find out the influence of different body condition to the pharmacokinetics of berberine,palmatine,baicalin,and geniposide and pharmacodynamics of anti-free radicalsResults:Macroporous resins combination 6(HPD100-AB-8 2:1)could refined Hunaglianjiedu Tang better and realize equivalent separation,unleash anti-cerebral ischemia effect.Pharmacokinetics of berberine,palmatine,baicalin,and geniposide demonstrate differences in the cerebral ischemia model andnormal rats.It shows that the pathological model rats had better absorption effect,and absorpt faster,have higher peak concentration than in the normal group.It has the same differences in SOD and MDA.The phenomenon indicates that cerebral ischemia may cause oxygen-poor injure in the body of rats After orally administered with Hunaglianjiedu Tang active fraction,SOD ascendes and MDA dsclineds,that shows Hunaglianjiedu Tang anti-cerebral ischemia active fraction can improve oxygen-poor injure.Conclusion:Macroporous resin combination combined with pharmaceutical screening is an effective way to gain Hunaglianjiedu Tang anti-cerebral ischemia active fraction;and we established the method to determine the content of berberine,palmatine,baicalin and geniposide in plasma,find out the differences of pharmacokinetics and pharmacodynamics in cerebral ischemia model and normal rats.It has made a new quest for research of pharmacokinetics and provide foundation for rational use of medicines and new drug manufacture about Hunaglianjiedu Tang.Besides,it is the first attempt to make use of macroporous resin combination to realize separation of multi-ingredients,and provide referential experiences for equivalent separation of Chinese herbal compound.
