Smilax scobinicaulis CH Wright belongs to the family Liliaceae,the rhizomes and roots of this plant have been used in folk medicine for the treatment of rheumatic arthritis,lumbago,gout,tumor and inflammatory diseases.Previous investigations of this plant show that flavonoid glycosides and steroidal saponins are rich in this plant,in order to obtain several cytotoxicity steroidal saponins we report the isolation and structure elucidation of flavonoid glycosides and steroidal saponins from the rhizomes and roots of S.scobinicaulis and cytotoxicity of steroidal saponins.The rhizomes and roots of S.scobnicaulis were extracted with 70%EtOH.The extract was suspended in H2O and partitioned with petroleum ether,EtOAc and n-BuOH,successively.The n-BuOH soluble fraction was subjected to D101 macroporous resin,silica gel CC,Sephadex LH-20,reversed phase silica gel CC and semi-preparative HPLC to afford 14 compounds.Then analysis of the UV?IR?MS?1H NMR?13C NMR and 2D-NMR spectrum of these compouds,6 flavonoid glycosides and 8 steroidal saponins were identified.Flavonoid glycosides were identified as hesperetin-7-O-[?-D-glucopyranosyl-(1?3)]-?-D-glucopyranosyl(1),clematine(2),ononin(3),daidzin(4),puerarin(5)and 3'-met hyloxy-puerarin(6),among them compound 1 was a new flavonoid glycosides and all of the six compounds were reported from the genus Smilax for the first time.Steroidal saponins were illuminated as 26-O-?-D-glucopyranoside-3?,26-dihydroxy-(25R)-5?-furostan-6-one-20(22)-en(7),26-O-?-D-glucopyranoside-3?,23,26-trihydroxy-(25 R,23R)-5?-furostan-6-one-20(22)-en(8),26-O-?-D-glucopyranoside-3?,26-dihydroxy-(25R)-5?-furostan-6-one-22-methoxyl(9),26-O-?-D-glucopyranoside-3?,26-dihydroxy-(25R)-5a-furostan-6-one-22-methoxyl-3-O-?-L-arabinopyranosyl-(1?6)-?-D-glucopyranoside(10),S milscobinosides A(11),(25R)-spirostane-5-en-3? 17a,27-trihydroxy-3-O-?-D-glucopyranos yl-(1?4)-[?-L-arabinopyranosyl-(1?6)]-?-D-glucopyranoside(12),(25R)-spirostane-5-en-3?,17?,27-trihydroxy-3-O-?-L-arabinopyranosyl-(1?6)-?-D-glucopyranoside(13),(25R)-sp irostane-3?-hydroxy-6-one-3-O-?-L-arabinopyranosyl-(1?6)-?-D-glucopyranoside(14),am ong them compound 7-10 were four new steroidal saponins.The cytotoxicity of compounds 7-10 were tested against human cervical carcinoma cell lines(Hela)and human hepatocellular carcinoma cell lines(SMMC-7221)by MTT method.The result revealed that the compound 13 displayed cytotoxicity against Hela carcinoma cell lines with IC50 values of 7.01 ±0.38 ?M and against SMMC-7221 cancer cell lines with IC50 values of 17.67±0.61 ?M,and compounds 10-12 and 14 showed moderate cytotoxic activity.
