This article summarized the physico-chemical properties and molecular structure of lentinan,the main purification methods and analysed the action mechanism of immunoregulation,antitumor,antivirus and antibacterial.The main conclusions were as follows:By studying the separation and purification of lentinan,an economic and efficient skill was established.1.The lentinan was graded and concentrated by membrane separation technology.Firstly,crude polysaccharide was divided into three parts by 500nm and 200nm ceramic membranes,which were named as ? 500nm part,200-500nm part and ? 200nm part.Then MWCO 10 K polyethersulfone membrane was used to concentrate the three parts.2.These fractions,separated by membrane,were further separated by HSCCC.Although the lentinan with high purity and uniform molecular were not obtained,it reduced the difficulty for the following gel purification.High-speed countercurrent chromatography was fit for separating the biological macromolecules such as polysaccharides.3.Len1 was separated by Sephacryl S100-400 gel filtration from the material which was separated by HSCCC.4.The contents(58.68%)of polysaccharides were determined with the phenol-sulfuric acid method.Separated by the membrane,HSCCC and gel chromatography,the total sugar content of Lenl was 95.3%.As no absorption at 260nm and 280 nm in the UV spectra,there were no nucleic acid and protein in these polysaccharides.Lenl was proved to be the homogeneous polysaccharide by the HPGPC chromatography.The method of separation and purification of lentinan is feasible and efficient,and laid a solid foundation for the follow-up analysis of the structure and anti-tumor activity of lentinan.
