Schisandra chinensis(Beiwuweizi in Chinese),a famous traditional Chinese medicine,grows wild in the north-eastern China(Jilin,Liaoning, Heilongjiang and so on)and is officially listed in the Chinese Pharmacopoeia.This plant has been revealed a widely variety of active effects including antihepatotoxic effect,antioxidant and detoxificant effect,anticarcinogenic effect,activity on central nervous system and counteract the fatigue,increase the durance,and improve the physical performance of sportsmen.The major active constituents of Beiwuweizi are considered to be lignans including Schisanhenol,Deoxyschisandrin,γ-schisandrin and Schisantherin A.Therefore,it is of important social significance and commercial values to investingate extraction of its useful compounts its exploition and utilization.With occurring the upsurge of savageness,nitrition and reture to nature all over the world,high-speed counter-current chromatography holds prominent ststus in the domain of preparative separation and purification of natureal products as a kind of effective and clean separation technology.The feasibility and the optimal condition of preparative separation and purification of lignans by high-speed counter-current chromatography from chisandra chinensis(Turcz.)Baill grown in liaoning province were investingated.Firstly,through an cleaning-up step on the silica gel,wo found abount 95%lignans were separated from the crude 95%ethanol extracts of chisandra chinensis(Turcz.)Baill with using petroleum ether-acetone as mobile phase.More,wo can analyzed the four lagnans,Schisanhenol Deoxyschisandrin,γ-schisandrin and Schisantherin A,by HPLC.Mobile phase was performed with methanol-water(75:25).The flow tate was 1.0ml/min.Detection wave was 254nm.Temperature was 35℃.Following an initial cleaning-up step on the silica gel,a preparative high-speed counter-current chromatography(HSCCC)with a two-phase solvent system composed of n-hexane-methanol-water(7:6:1,v/v)was used to isolate and separate Schisanhenol and Deoxyschisandrin,γ-schisandrin and Schisantherin A from Schisandra chinensis with the upper organic phaes system as the stationary phase and the lower phase system as the mobile phase.A total of 122mg Schisanhenol and 86 mg Deoxyschisandrin and 93 mgγ-schisandrin.Especically the Schisanhenol was separated at the first time in this way.Compare with the 2 compounts before,we have gotten 4 active lignans after preparative separation and purification by high-speed counter-current chromatography once.Since we uesd the 95%ethanol as the extraction agent which can disslove more kinds of lignans.So,we can raise the materials use factor.
