| Modern research has found that the antitumor active ingredient in lycoris radiata plant is mainly alkaloids.Crinum asiaticum is one of the representatives of lycoris radiate plant.It contains very rich alkaloid composition and fairly extensive pharmacological effects,such as anti-malarial,to stop sputum,anti-arrhythmic,antihypertensive,emetic effect and other effects.Now the research of Crinum asiaticum of antitumor active ingredient is not deep,therefore it is necessary to analyze Crinum asiaticum at all aspects,this experiment regards strong alkaline alkaloids in Crinum asiaticum alkaloids as the research object,through the separation,purification and LC-MS technology,it is speculated that possible strong alkaline component in Crinum asiaticum extractum,it provides powerful experimental basis and reference data for clinical use of Crinum asiaticum alkaloids.Experimental process is divided into three parts.First,strong alkaline extraction process of Crinum asiaticum L.alkaloids is optimized.Then the extraction of strong alkaloids from Crinum asiaticum L.alkaloids is purified and separated and it is analyzed by liquid chromatography.Tinally,the extraction of strong alkaloids from Crinum asiaticum L.alkaloids is analyzed by mass spectrum.It is speculated that there may be exist alkaloids types.The experiment is running by the classical extraction method,using ethanol to exact the alkaloids of Crinum asiaticum L.seed,emoving impurity by using the method of acid soluble alkali sink,the strong base alkaloid can be obtained.First of all,ethyl acetate is used to extract the low alkaline alkaloid ingredients,then the extract is obtained by concentrating aqueous alkali.By using thin layer chromatography,color developing agent and ultraviolet light shines,the alkaloids can be found in the sample.Then the different polarity of strong alkaline alkaloids can be got by the adsorption of column chromatography on silica gel.The alkaloids will be collected and purified by using the SPE-C18.Finally,through the analysis of LC-MS,it can be inferred that the size of the molecular weight of the strong alkaline alkaloid and further can be analyzed its structure characteristics.Through the orthogonal experiment analysis,the best extraction technology for total alkaloid ultimately is determined.It is ethanol concentration 85%,extraction temperature is 65℃,extraction time 3 h.The liquid ratio is 8:1 and extraction times are 3.By comparison,the effect of alumina thin-layer chromatography is better than silica gel thin-layer chromatography.When developing solvent is water and methyl alchol and ratio is one to two,two alkaloids color points can be obtained and the points are with fluorescent effect.The purified and concentrated eluent of silica gel column is analyzed by LC-MS technology.Initial speculation component may contain six active ingredients.That is Pseudolycorine,N-Methyldendroblum,Ungeremine,Lycoramine,Columbin and Lycorine.Hope that through further infrared spectrum(IR)and nuclear magnetic resonance(NMR)method to determine the strong alkaline alkaloid structure characteristics in order to provide theoretical basis for the strong alkaline alkaloid pharmacological studying. |
