Objectives:Using in vitro bacteriostasis experiment study bacteriostatic effect of liposoluble constituent components and water-soluble components in Cnidium monnieri.Using the compound condensing method liposoluble constituent components of Cnidium monnieri preparation into microcapsule,using cold win min gel was prepared,and the common cnidium fniit millirod win min gel quality standards.Methods:This paper adopts high performance liquid chromatograpHy determination of index components in cnidium monnieri common cnidium fruit content,in order to extract and cnidium monnieri content as an index,select the best extracting fructus cnidii fat-soluble method,with L9(34)orthogonal test to optimize the extraction process of cnidium monnieri liposoluble constituent components.Such as coating rate,drug-polymer interactions as an index,select the best cnidium monnieri microcapsule preparation method,and the single factor investigation capsule material concentration,curing agent concentration,into a capsule in the process of the influence of temperature,pH,stirring speed on the preparation.L9(34)orthogonal test make optimize cnidium monnieri preparation technology of the microcapsule.Gelling temperature and release as indexes,the single factor to investigate the preparation of Cnidiun monnieri Temperature sensitive microcapsule gel prescription factors influence.Bacteriostatic circle by measuring the size of cnidium monnieri fat-soluble composition and water-soluble components bacteriostatic experiment was carried out.According to ChinesepHarmacopoeia method using plasma part two determination of Cnidium monnieri temperature sensitive microcapsule gel deggree of release.Results:With ethanol reflux method to extract fat-soluble components in fructus cnidii,orthogonal test table to determine the best extraction technology for six times the amount of 95%ethanol,extracting time is 2h.Cnidium monnieri fat soluble and water-soluble components of bacteriostasis experiment results show that both parts composition has inhibitory effection on different bacteria,and fructus cnidii fat-soluble components inhibitory effect is extremely obvious.The compound condensing of cnidium monnieri prepared by micro capsule,the single factor investigation microcapsule material concentration was 5%,curing agent,glutaraldehyde concentration 20%,emulsifying method to organize the mash,curing time of 1.5 h.L9(34)orthogonal test to determine the best micro capsule preparation technology for:encystment temperature is 50 ?,pH of 4,stirring speed of 600 r/min.From cnidium monnieri micro capsule coating rate was 78.22%,the drug loadings was 14.16%,the average particle size of around 12 microns.According to the single factor investigation and L9(34)orthogonal test,determine fructus cnidii WenMinWei capsule prescription gel ratio for P407:P188:propylene glycol = 18%:1%:15%.By thin layer chromatograpHy(TLC)team cnidium monnieri WenMinWei capsule gel by qualitative identification of fructus cnidii,the results showed that with the reference substance and contrast medicinal materials of the same level,and the dark blue color remains the same point.By high performance liquid chromatograpHy(HPLC)determination of cnidium monnieri content,and tentatively cnidium monnieri WenMinWei capsule cnidium monnieri element content in the gel should be not less than 31.77?g/mL gelling temperature should be within 36.0 ? to 37 ?.By the in vitro release,ccording to the results of cnidium monnieri drug release completely within 0.5 h,ommon cnidium fruit gel as gel dissolution and make the drug release,rug release within 180 min 89.12%common cnidium fruit gel has certain slow-release effect.hen cnidium monnieri fat-soluble components released from cnidium monnieri drug release speed,ay be due to the gel matrix poirot sharm for non-ionic surface active agent,peed up the dissolution of refractory material.And fructus cnidii WenMinWei capsule gel in the release process slows down the cnidium monnieri fat-soluble components release rate,xtend the time of the release of drug,reached the slow-release effect.Conclusion:Active ingredients of Cnidium monnieri mainly exist in the liposoluble constituent components,common cnidium fruit micro capsule preparation process is stable.Cnidium monnieri micro capsule temperature sensitive gel which is formed by the slow release effect is good,stable quality standards,application prospect.
