Objective Flavonoids play an important role in Cadiovascular protection and flavonoids in Carya cathayensis leaf is also a kind of Flavonoids.The method of Intragastric administration was used to study the effect of flavonoids in Carya cathayensis leaf on mice bleeding time and coagulation time,myocardial ischemia in mice,lowering blood pressure of spontaneously hypertensive rats and lowering blood lipids in mice hyperlipemia model in order to explore the protective effects of flavonoids on the cardiovascular system.Methods(1)The preparation of flavonoids in Carya cathayensis leaf.Extracting solution will obtained by the method of reflux extraction.And then mixed with polyamide,removed the water by reduced pressure distillation.chromatographed on column and eluted with 30%ethanol.After eluted with 40%ethanol,collected the eluent,removed the water by reduced pressure distillation.Finally,High performance liquid will be used to determine the content of flavonoids in the collected product.(2)Effects of flavonoids on the bleeding time and coagulation time in mice.The ICR mice will be randomly divided into 4 groups by body weight.Normal control group,low dose group(50.0 mg/kg),medium dose group(100.0 mg/kg)and high dose group(200.0 mg/kg).The number of each grou is 10.The method of Intragastric administration will be used for 16 days.In 16 days,1 hour later after Intragastric administration,cut the end of mice tail and record the time.At the same time,use the capillary tube to obtain Orbital blood,and drop of blood in new slides.Use a needle to turn the blood gently,to observe the phenomenon,and record the clotting time.(3)Effects of flavonoids against myocardial ischemia in mice.The ICR mice will be randomly divided into 5 groups by body weight.Normal control group,Model control group,positive control group(propranolol 20.0mg/kg),low dose group(100.0 mg/kg)and high dose group(200.0 mg/kg).The number of each group is 10.The method of Intragastric administration will be used for 30 days,and the intragastric volume was 0.1 ml/10g weight.In 28 days and 29 days,30 mg/kg of isoprenaline hydrochloride will be subcutaneous injection.Weighted the body weight of mice,collected the blood,centrifugal and taken supernatant.Determined the content of LDH and CK in blood,dissection,taken heart and weighted the weight of the heart.Then heart will baked under the conditions of 110 ℃ for 8 hours,weighted and calculated the cardiac index and the water content of heart muscle.(4)Effects of flavonoids to lower blood pressure of the spontaneously hypertensive rats.Based on the underlying value of blood pressure,the SHR were rando mly divided into 5 groups.Model control group,Captopril group(15mg/kg),low dose group(50.0 mg/kg),medium dose group(100.0 mg/kg)and high dose group(200.0 mg/kg).The number ofeach group is 10.The method of Intragastric administration will be used for 31 days.Observed and measured the changes of SBP,DBP and MBP of rat blood every other week.(5)Effects of total flavonoids to the influence of hyperlipemia mice.Female ICR mice were administrated with high-fat forage in 23 days in order to build hyperlipemia model.After 23 days,these mice were divided into model group,low-dose group(100.0mg/kg)and hight-dose group(200.0mg/kg)with the feeding high-fat forage in process of total experience.Control group were fed with common forage in process of total experience.Low-dose group(100.0mg/kg)and high-dose group(200.0mg/kg)were administrated with total flavonoids from Carya cathayensis leaf,while model group and Control group were administrated with 3%tween-80 solution in 60 days.Mice in different groups were killed after the last drug administration.TC,TG,HDL-C,LDL-C,AST,ALT,UA,CR were detected in serum Livers were strained with HE in order to observe their morphological change.Results(1)The preparation of flavonoids in Carya cathayensis leaf.The five known flavonoids in the totalcontent are above 50%.They are Chrysin,Wogonin,Cardamonin,Pinostrobin and Pinocembrin.This was consistent with the experimental expectations.(2)Effects of flavonoids on the bleeding time and coagulation time in mice.The content of flavonoids in 50.0mg/kg,100.0mg/kg and 200.0mg/kg were able to prolong the bleeding time and clotting time in mice,which provides a basis for Antithrombotic about flavonoids.(3)Effects of flavonoids against myocardial ischemia in mice.The content of flavonoids in 100.0mg/kg and 200.0mg/kg were able to lower CK in myocardial ischemia mice significantly.And the content of flavonoids in 200.0mg/kg was able to lower LDH in myocardial ischemia mice to a certain degree.However,the content of flavonoids in 100mg/kg and 200.Omg/kg could not reduce myocardial water content in myocardial ischemia mice by reducing the degree of cell swelling.(4)Effects of flavonoids to lower blood pressure of the spontaneously hypertensive rats.The content of flavonoids in 50.0mg/kg,100.0mg/kg and 200.0mg/kg were able to lower systolic pressure of SHR on an varying degree.In addition,the content of flavonoids in 100.0mg/kg and 200.0mg/kg were able to lower diastolic pressure and diastolic pressure of SHR.(5)Effects of total flavonoids to the influence of hyperlipemia mice.After building the model of hyperlipemia mice,mice were administrated in total flavonoids(100.0mg/kg)and(200.0mg/kg)in 60 days and their TC,TG,LDL-C were higher than model groups’,while their HDL-C were lower model groups’.But livers and renals from groups administrated total flavonoids were not dysfunctioned by total flavonoids and were protected from total flavonoids.Conclusion The five known flavonoids in the total content are above 50%.By continued pharmacological experiments,we found that,flavonoids had the effect of prolonging the bleeding time and clbtting time in mice,against myocardial ischemia in mice,lowering blood pressure of the spontaneously hypertensive rats and enjoying the protective function of livers.But total flavonoids could not lowering the blood lipids in mice hyperlpemia model As a result,total flavonoids in Carya cathayens is leaf could protect the cardiovascular system preferably.
