She Liu Gu,which is also called Konjac,is a perennial herbaceous plant of araceae.The tuber contains abundant konjak glucomannan,starch,protein,mineral substalce,alkaloid,et.[1-5]Konjac possesses special nature and pharmacology functiona,and it has very good application values on anti-tumor,lipid metabolism,glycometabolism and enhancing immunity.It is a study on extraction and purification and structure-activity relationship,thus,providing theoretical basis for its development and utilization further.The followings are the main research contents.(1)In order to achieve the best method of konjac alkaloid's extraction,spectrophotometry detecting and orthogonal experiment were taken to study the best method of konjac alkaloid's extraction in ultrasonic assisted extraction,ethanol extraction,soxhlet extraction and microwave extraction respectively.The result showed that the best conditions of ultrasonic assisted extraction are 75%ethanol concentration,1:20 solid-liquid ratio,40 minutes and 80? for extraction,and the yield of alkaloids was 0.484%.The best conditions of ethanol extraction are 75%ethanol concentration,1:30 solid-liquid ratio,four hours and 70oC for extraction,and the yield of alkaloids was 0.432%.The best conditions of soxhlet extraction are 85%ethanol concentration,1:20 solid-liquid ratio,four hours for extraction,and the yield of alkaloids was 0.482%.The best conditions of microwave extraction are 75%ethanol concentration,1:30 solid-liquid ratio,30 minutes for microwave time,and the microwave power is 600w.The yield of alkaloids was 0.432%.These four extraction methods were compared with each other,and it turned out that ultrasonic assisted extraction is more suiTab for industrial production with its low production cost,simple proeess and high extraction rate.(2)The study focused on the process of D101 macroporous adsorption resin to purify konjac alkaloid.The method is to use spectrophotometry detecting to survey the sample quantity,adsorption capacity,and the concentration and volume of ethanol on D101 macroporous adsorption resin.The results showed the purification process:the sample quantity of D101 macroporous adsorption resin is 50ml;The sample rate is lml/min.The best result is at the condition of eluting by 90%ethanol,and 50ml eluent.At this condition,the effect of primary separation is good.(3)Using the method of topological index to predict the structural properties.A series of imidazole derivatives which processing biological activity were studied on structure-activity relationship to build molecular connectivity index.This study focused on the relevance between the physicochemical property of imidazole derivatives,biological activity and molecular connectivity index.The results showed that as the increasing of imidazole derivatives' substituent amount,the atomic point valence of substituent is enlarged;the chain is lengthened;the branching degree of substituent is enhanced.The researchers also found a positive correlation between molecular connectivity index and it.Therefore,molecular connectivity index has good structure selectivity and biological activity relevency,and it is available for predict the activity of imidazole derivatives.
