This experiment aims to study of astragalus polysaccharide containing selenium on inhibition of human gastric cancer SGC-7901 cells and explore the mechanism and approach through the related experiments.In the experiment,MTT results showed that astragalus polysaccharide containing selenium has certain inhibitory effect on the growth of SGC-7901 cells,and with the increasing concentration of astragalus polysaccharide containing selenium the inhibition present dose correlation.Through data analysis,the result of IC50 is 77.5?g/mlTo observe the direct effects of astragalus polysaccharide containing selenium in human gastric cancer SGC-7901 cells by inverted microscope after 48h,cells of negative control group a compared with cells of drug group are arranged in relatively close and morphological integrity.With increasing concentration,cell density of drug groups become thin from dense;Cells shrank and stick reduced.The part of cells in high dose group has been broken and the morphology is not complete.The cells in the experimental group impacted by Astragalus polysaccharide containing selenium after 48h are observed under fluorescence microscope after Hoechst33258 staining.The negative control group cells nuclei showed diffuse homogeneous blue fluorescence and morphological rules.Low dose group nuclear morphological changes and become pyknosis.Medium dosage group presents nuclear fragmentation,The high dosage group nuclei were condensed and the apoptotic bodies appeared.The two experimental data showed that astragalus polysaccharide containing selenium can lead SGC-7901 cells into change from morphology.And the preliminary forecast may be that the inhibition of cell growth was due to the cell apoptosis.The next experiment,we use propidium iodide(PI)staining SGC-7901 cells effected by different concentration of astragalus polysaccharide containing selenium after 48h.The results found that the appearance of apoptosis peak by flow cytometry.And the data analysis of each group cells apoptosis rates were(1.15±0.22)%,(4.75±0.32)%and(19.06 ± 0.75)%,like dose dependent.We determined the inhibition to SGC-7901 cells is associated with induction of apoptosis of tumor cells in the action of astragalus polysaccharide containing selenium.Through the above experimental results as the basis,the next experiments we also investigated the apoptosis of human gastric cancer cell line SGC-7901 induced by astragalus polysaccharide containing selenium.SGC-7901 cells effected by different concentration of astragalus polysaccharide containing selenium after 48h were observed under the laser scanning confocal microscopy.We could find the changes in the mitochondrial membrane potential,the content of active oxygen and calcium ion concentration.The fluorescent intensity of mitochondrial membrane potential were 32.83±1.21;27.42±0.82;17.91±0.70,fluorescent intensity of ROS were:17.63±1.14,43.97±4.07,113.80±6.79 and the fluorescence intensity of Calcium ion were:9.10±0.06,11.27±0.07,23.59±0.42.The data of each group compared with the control group were statistically significant(P<0.05).The above results show that,the mitochondrial membranecell of the cells effected by astragalus polysaccharide containing selenium were destroyed with the increase administration.The mitochondrial membrane potential was decreased,the content of ROS and calcium ion increased,those result in the MPTP opening,finally lead the cells to apoptosis.To sum up,in this paper,we can prove that astragalus polysaccharide containing selenium could induce apoptosis of human gastric cancer SGC-7901 cell by vitro experiments.And the mitochondrial pathway is one of the possible ways of apoptosis of human gastric cancer SGC-7901 cells induced by astragalus polysaccharide containing selenium.
