| Early sale of traditional Chinese medicines is the pattern of "front shop,back mill".There was no clear boundary between primary processing and processing crude drugs.The two processing were collectively referred to as processing crude drugs.With the development of traditional Chinese medicine(TCM)commercialization,primary processing and processing crude drugs are separated into two independent processing systems artificially to make convenient for the carriage and long-term storage.Although the separation improves productivity,it ignores the inherent law and relevance of drugs quality formation,gradually exposing shortcomings.Nowadays The establishment of the GMP base and GAP base provides the integrated processing technology foundation.Therefore,the thesis selected Chinese medicines that with solid research foundation and had certain representativeness in medicinal parts,primary processing and processing crude drugs as study object to elucidate the rationality of the integration processing technology based on the comparison on chemical constituents and pharmacological effect between traditional processing and integration processing.The traditional processing was based on the《Chinese Pharmacopoeia》(2010,2015)and primary processing and processing crude drugs in geo-authentic habitats.Chapterl.Literature researchIn this section,the chemical constituent,pharmacological effects,primary processing and processing crude drugs of Chaenomelis Fructus,Gastrodiae Rhizoma,Schizonepetae Herba,Paeoniae Radix Alba and Rehmanniae Gadix were systemtically reviwed.The latest development of material basic and pharmacological effects in recent years were summarized and analyzed to confirmed the main efficacy components and main functions.Based on summarizing the experience and achievements of the study on traditional processing,and the influence of modern processing technology of traditional Chinese medicine,the thesis thoughts,the integration processing,were obtained and reasonable and feasible experimental programs and technical courses were designed.Chapter2.The Study on the integration processing of Chaenomelis FructusThe content of oleanolic acid and ursolic acid were regarded as the study indicators to investigate the appropriate integration processing technology that was heating up in hot water for 5 minutes,steaming for 4 minutes and drying for 5 hours at 60℃.The experimental result of pilot scale verified the Stability and feasibility of the appropriate integration processing technology.And the content of organic acid of Chaenomelis Fructus produced from the integration processing was higher than from the traditional processing.Compared with the model group,it was shown to exert significant inhibitory effect on secondary reaction of adjuvant-induced arthritis of rats in the integration processing group and the traditional processing group,that lowering the degree of paw swelling and arthritis index,decreasing the activity of TNF-α,IL-1β,increasing the concentration of IL-10.However,the integration processed products method was more effective at high doses.The integration processing ensures the quality,reduces the cost of production and improves production efficiency,being feasible and reasonable.Chapter3.The Study on the integration processing of Gastrodiae RhizomaOptimum integration process was as follows:Gastrodiae Rhizoma was steamed for 30min,and dried for 12h at 60℃.The experimental result of pilot scale verified the Stability and feasibility of the appropriate integration processing technology.The contents of gastrodin and parishins of Gastrodiae Rhizoma produced in the integration processing were higher.The effects of the product in the model of PTZ induced eclampsia were observed to compare the product in the different processing technology.Both the integration processing group and the traditional processing group could relieve convulsion and reduce the death of mice.But at high dose,the anti-eclampsia effect of Gastrodiae rhizoma produced in integrated processing technology was better since the longer.incubation period of paroxysmal spasm and tonic convulsion and greater protection against death.Compared to traditional processing technology,integrated processing technology is better in guaranteeing the quality of pieces,reducing the cost of production and increasing the production efficiency.The rationalization of the integrated processing technology is expounded by the comparison on the product in different processing methodsChapter4.The Study on the integration processing of Schizonepetae HerbaThe content of volatile oil was regarded as the study indicators.The appropriate integrated processing technology was as follows:dry at 50℃ for 5h,cut into segments,dry at 50℃ for 3h.The experimental result of pilot scale verified the Stability and feasibility of the appropriate integration processing technology.Results showed that the contents of chemical components in the products obtained with the integrated processing were higher than those with the other processing.And both the Schizonepetae produced in the two processing could reduce the degree of swelling and the content of TNF-α/IL-1β/IL-6 in the serum.Compared with the traditional processing,the integration processing technology ensures the quality of the decocting pieces,reducing the cost of production and improving production efficiency.Chapter5.The Study on the integration processing of Paeoniae Radix AlbaOptimum integration process was as follows:Paeoniae Radix Alba was boiled for 15min,and dried for 6h at 50℃.The experimental result of pilot scale verified the Stability and feasibility of the appropriate integration processing technology.The effects of the product in the model of hemorrhagic blood-insufficiency were observed to compare the product in the different processing technology.Compared with the model group,the level of RBC,HGB,HCT and PLT in the the traditional processing and the integration processing group were remarkably higher.The spleen index was lower,while the thymus index was increased.The activity of Na+-K+-ATPase and Ca2+-Mg2+-ATPase of RBC in the two processing were all obviously enhanced.Both the Paeoniae Radix Alba produced in the two processing can enhance the hematopoietic function and energy metabolism of RBC.The integration processing not only guarante the quality of the decocting pieces,but also reduce production cost and improve production efficiency compared with the traditional processing.Chapter6.The Study on the integration processing of Rehmanniae GadixThe appropriate integrated processing technology was follows:the drying temperature was 50℃,and the drying time was 18 hours.The experimental result of pilot scale verified the Stability and feasibility of the appropriate integration processing technology.The contents of catalpol and acteoside were 4.31%for catalpol and 0.31%for acteoside in the integrative processed samples;the corresponding results in traditional processed samples were 1.25%and 0.04%,respectively.Rehmanniae Radix processed by the both integrative and traditional processing techniques showed the obviously therapeutic effects in rats,which were administered with the dried ginger extract by oral gavage daily to induce the blood-heat and hemorrhage syndrome model,based on the measurement results of body temperature,RBC,the blood viscosity and plasma viscosity,thrombocytes and coagulation function.And no significant differences were found between two different processing techniques with respect to the efficacy of Rehmanniae Radix products.In conclusion,the integration processing can replace traditional processing to process Chinese medicine.The integration processing has certain feasibility and rationality. |
