| Objective: To explore the expression of Bruton’s tyrosine kinase(Btk)in the process of periapical lesions in human,analyze the relation of Btk and bone resorption in periapical periodontitis,and discuss the mechanism of bone resorption.Methods: Twenty-five specimens of human chronic periarthritis tissues and nine healthy tissues were collected with the consent of the patients.Divided into two groups,one used the method of Real-time PCR which extracted m RNA from fifteen periarthritis tissues and six healthy ones to detect the expression of Btk m RNA;the other group used the method of Western Blot which extracted protein from ten periarthritis tissues and three healthy ones to detect the expression of Btk protein.Images and data were collected for statistical analysis.Results: The expression of Btk is much higher in human chronic periarthritis tissues were obviously higher than the healthy ones.The results of Real-time PCR showed that,six healthy tissues were detected a small amount of Btk m RNA and only three ones are positive,however Btk m RNA expressed in all periarthritis tissues and much higher the healthy ones,the difference was statistically significant(P<0.05).Western Blot results showed that Btk protein was much higher in human chronic periarthritis tissues were higher than the healthy ones,with statistical significance(P<0.05).Conclusion:The expression of Btk is higher in periapical periodontitis,which reminded that it played an important role in the progression of periapical periodontitis,so we speculate that Btk is involved in development of chronic periapical periodontitis and the absorption of bone tissue. |
