| Objective:The purpose of this study was to collect Hunan People’s Hospital esophageal carcinoma tissues,para cancer tissues and Barrett esophageal tissue samples by immunohistochemistry,Western blot and other molecular biology means to carry out the study of the CTGF expression,the relevant factors for esophageal carcinoma and the expression of CTGF.Objective to explore the related factors of CTGF expression in esophageal carcinoma,and to explore the mechanism of CTGF involved in the occurrence and development of esophageal carcinoma.Methods:In this study,according to the needs,classification statistics of age and sex,histological classification,esophageal cancer TNM staging,pathological types of esophageal cancer,lymph node metastasisand distant metastasis situation were done.Expression analysis by means of immunohistochemistry and Western blot blot was used to evaluate CTGF protein in these tissue samples.According to the standard statistical expression of positive expression,the positiveand negative rate of protein expression were compared in different age,gender,esophageal cancer the organization,TNM staging,the pathological types of esophageal cancer,lymph node metastasis,and distant metastasis situation.By using univariate and multivariate regression analysis to explore the factors influencing the expression of CTGF in esophageal cancer tissue samples.Further using the same methods,comparison of CTGF expression in esophageal cancer tissue samples,Barrett esophagus and in adjacent normal tissues were done.which suggested that CTGF value and evaluation in predicting the occurrence and development of esophageal cancer and metastasis,angiogenesis mechanism,to provide basis for improving the diagnosis and treatment of esophageal cancer clinical new target.Results:1、The difference of the expression of CTGF in esophageal carcinoma tissues of different gender and age was compared.The results showed that the comparison of.positive expression rate of CTGF in male and female patients with esophageal carcinoma was not statistically(p>0.05).Compared with the [30 group,60 years old),the positive expression rate of CTGF in [60 group,80years old)was not statistically significant(p>0.05).2 、 The credit of different kinds of tumor tissue like esophageal cancer tissue samples CTGF difference comparison results show that compared with squamous cell carcinoma tissue samples,the positive expression of CTGF in adenocarcinoma was significantly increased(p<0.01)with significant statistical difference.3、According to CTGF expression comparison results in the different TNM staging of patients with esophageal cancer tissue samples,compared with the esophageal cancer tissue samples in phase 0,the positive expression rate of CTGF in esophageal cancer tissue samples in phase II B,III,IV,IV,IV A was significantly increased(p<0.01)with significant statistical difference.Compared with the esophageal cancer tissue samples in phase 0,the positive expression rate of CTGF in phase I,phase II A was not statistically significant(p>0.05).4 、 According to CTGF expression comparison results in 4different esophageal cancer pathological tissue samples,compared with the hidden type of esophageal cancer samples,the positive expression of CTGF in the esophageal cancer tissue samples was not statistically significant(P>0.05).5、According to CTGF expression comparison results between withand without lymph node metastasis of esophageal cancer tissue samples,compared with no lymph node metastasis in esophageal carcinoma tissue samples,CTGF positive expression in metastatic lymph node samples of esophageal carcinoma tissue was significantly increased(p<0.01)with significant statistical difference.6、According to CTGF expression comparison results between with and without distant metastasis in esophageal cancer tissue samples,with no metastasis of esophageal cancer tissue samples,CTGF positive expression in distant metastasis of esophageal carcinoma tissue was significantly increased(p<0.01)with significant statistical difference.7、By univariate analysis and Logistic regression analysis,the classification,tumor TNM staging,lymph node metastasis and distant metastasis were proved to be the related factors affecting the expression of CTGF in esophageal cancer tissue samples.8 、 According to CTGF expression comparison results in esophageal cancer,esophageal Barrett and esophageal cancer tissues,compared with adjacent normal tissue samples,the positive expression rate of CTGF in Barrett esophageal tissue samples increased significantly(p<0.05)with significantstatistical difference,and the positive expression rate of CTGF in adenocarcinoma,squamous cell carcinoma and esophageal cancer(adenocarcinoma and squamous cell carcinoma)tissue samples increased significantly(p<0.01)with significant statistical difference.Conclusions:The main conclusions are as follows:1.The expression of CTGF in esophageal cancer tissues was not correlated with the age and sex of the patients.2.Through univariate and multivariate regression analysis,the histological classification,TNM stage,lymph node metastasis,and distant metastasis were the factors that influence the expression of CTGF in the tissues of esophageal carcinoma.3.Compared with para cancer tissues,CTGF was highly expressed in esophageal carcinoma.in esophageal carcinoma,and the expression of CTGF was up-regulated in Barrett esophageal.4,This result also further suggests that CTGF may regulate the angiogenesis of esophageal cancer involved in the formation of esophageal carcinoma. |
