Objective:To investigate the effects of RNPC1 gene on the proliferation,migration and invasion of malignant melanoma cells A375.Methods:RNPC1 gene over-expression(group O)and short hairpin RNA interferencing RNPC1 gene expression(group L1 and L2)were built in malignant melanoma cells A375.The blank control group(group ON)and negative control group(group LN)were built as well.The expressions of RNPC1 mRNA and protein were detected by real-time fluorescent quantitative PCR and Western blot,respectively.The proliferation,colony-formation,migration,and invasion of malignant melanoma cells A375 were measured by CCK-8,wound healing assay,plate colony assay and Transwell assay.Results:The expressions of RNPC1 mRNA and protein were lower in group L1 and L2 than those in Group LN,which were higher in group O than those in group ON(P<0.01).The capability of proliferation was weaker in group L1 and L2 than that in group LN,which was stronger in group O than that in group ON(P<0.01).The capability of colony-formation was weaker in group L1 and L2 than that in group LN,which was stronger in group O than that in group ON(P<0.01).The capabilities of cell migration and invasion were weaker in group L1 and L2 than those in group LN,which were stronger in group O than those in Group ON(P<0.01).Conclusion:RNPC1 gene over-expression can strengthen the proliferation,colony-formation,migration and invasion of malignant melanoma cells A375,which may provide a new target for clinical Treatment.
