Objective:To study the effect of different concentrations of sodium fluoride on proliferative activity of the human malignant melanoma cell line A375(A375 cell line) in vitro and explore the treatment of NaF on human malignant melanoma (MM). Methods:After cells were cultured with various concentrations (10,100,500,1000,5000,10000,20000 and 40000umol/L) sodium fluoride for 24 hours and 48 hours, the proliferation of A375 cell line was measured by MTT method. The morphological changes of the cell were observed under Inverted microscope. Results:NaF promoted the proliferation of melanoma cell when it was 10umol/L. Started from 100umol/L, the survival rate of melanoma cells reduced and the inhibition rate enhanced along with increased drug concentration and time extension. However,10000,20000 and 40000umol/L effected on A375 cells for 24hs, 20000 and 40000umol/L for 48hs all had no difference(p>0.05).100 and 500umol/L showed an inhibitory effect. If the concentration of NaF was above 500umol/L, cytotoxicity appeared:the number of cells significantly decreased,cell detached,changed rounded and smaller, nucleus disappeared and so on. Conclusions:Fluoride acts directly on A375 cell line to stimulate cell proliferation in a biphasic fashion, namely inhibits proliferation,even directly kill cells at high concentration, while promoting proliferation at low dose.The inhibitory rate is related with Sodium Fluoride concentration and incubation time.
