Objective:1)To Study optimum extraction condition of Cistanche salsa for laxative extract.2)To study laxative activities and safety evaluation of laxative extract by animal and microorganism experimental,to provide the basis for development of Cistanche salsa laxative products.Methods:1)Use the extract yield and content of dulcitol as parameters,using single factor and orthogonal experiments to optimize the extraction process.2)To conduct qualitative and quantitative detection on laxative extract by TLC and UV-vis.3)Establish constipation mice model by using diphenoxylate tablets,the small intestine motor and relieving constipation experiments were used to investigate laxative effect of extract.4)Investigate changes of intestinal flora in mice feces after treatment with extract,by observing the changes of four kinds of microbial flora,escherichia coli,salmonella,lactobacillus and Bifidobacterium.5)The acute toxicity and 30 days feeding test were used to evaluate the safety of laxative extrct from Cistanche salsa.Results:1)The optimum extraction condition of Cistanche salsa laxative extract were:Ethanol concentration was 40%,material liquid ratio was 1:16,extracting time was 2.0 h and for three times.2)TLC conditions for Dulcitol of laxative extract was:developing agent is ethyl acetate:isopropanol:water:formic acid?3:4:2:1?with 3%acidic potassium permanganate as chromogenic reagent.TLC conditions of Betaine was Methanol:Water:Formic acid?8:2:1?developing with modified Drogendoff's as chromogenic reagent.TLC conditions of Allantoin was Ethyl acetate:Acetone:Formic acid?8:1:3?developing with25%p-Dimethylaminobenzaldehyde as chromogenic reagent.Sugar alcohols content was determined by appling UV-vis spectrophotometry,and the content was 20.1%used Dulcitol as standard reference.3)Compared to control group,different concentration aqueous solution of Cistanche salsa laxative extract can obviously shorten the first defecation time?P<0.01?,it can also increase dejection amounts,weights,and charcoal propulsion rate in 6 hours?P<0.01?.4)Cistanche salsa laxative extract can promote proliferation of the intestinal probiotics and Bifidobacterium,however,it can inhibit the Escherichia coli.5)For it was unable to get LD50 by preliminary acute toxicity experiment,the maximum dosage method was adopted and maximum tolerance dose is104 g/kg·bw.There isn't any obvious adverse reaction to rat body weight,blood and the major organs in30 days feeding experiment.Conclusion:1)The extraction process of Cistanche salsalaxative extract and the qualitative and quantitative analysis method of Dulcitol is reasonable,easy operation,economy,and is suitable for mass production industrialization.2)Cistanche salsaextract can promote intestinal peristalsis,regulating intestinal flora with runchang purge efficacy.3)Cistanche salsa as health care products for human use is safe and non-toxic.
