Objective:1.To scientifically and rationally revise the commodity specification grade standards of Salvia miltiorrhiza,in order to regulate the trade of Salvia miltiorrhiza,and contribute to providing the basis on good quality and reasonable price.2.Key elements of medicines specification comprise of production area,processing methods,as well as drying methods.A reasonable and comprehensive quality assessment of Salvia miltiorrhiza.Considering the multi-component analysis systematically throughout every process supports the scientific and reasonable evidence of classification.3.Studying on different levels of characters and internal components on changing rules of Salvia miltiorrhiza in current production areas and markets,which will ensure the variety of grades.Methods:1.Conducting investigations in production areas and markets.1.1 Surveys of production areas on Shandong,Sichuan,Anhui,Shanxi,Hebei,Shaanxi and Henan are to understand the production processing,grading conditions of production areas,and collect samples.1.2 Carrying out commodity surveys and samplings on Bozhou,Anguo,Hehuachi and Yulin these four large Chinese herbal medicine markets are needed.1.3 Exploring the factors affect the quality of Salvia miltiorrhiza in the production areas and markets of each link.1.4 Data measurement and regularity analysis of the characters of Salvia miltiorrhiza are going to be conducted,summarizing the methods of division in different grades.2.Liquid-mass spectrometry technology was adapted to study the cracking rules of tanshinones and salvianolic acids,and systematically identifies the components of Salvia miltiorrhiza.Differentiate the grades,production areas,processing methods and drying techniques was to consider various components.The change of chemicalcomposition before and after mildew of Salvia miltiorrhiza was analyzed3.Simultaneous determination of indicative components between 4 salvianolic acids and 3 tanshinones in Salvia miltiorrhiza had accomplished by using UPLC.4.Measuring polysaccharides contents of Salvia miltiorrhiza samples among different grades and origins were undertaken in the way of anthrone-sulfuric acid method,meanwhile the data correlation analysis were proceeded by statistics method.5.Sequencing technology were adapted to appraise relevant molds.Results:1.The current specifications of Salvia miltiorrhiza all over the place have been divided separately,in which Zhongjiang Danshen from Sichuan has the most detailed division.According to the size of diameter,it consists of 4 to 5 grades.Briefly division has been carried out in Shandong producing regions.Besides,other growing districts are not graded,and less original herbs are showing on the market.Moreover,most of medicines are cut into segments or slices,and sifted into range of sizes.There are also specifications of sulfur and diaphoretic to various degrees existing on markets,and specification divided in an informal way.2.Different producing regions are influenced by different habitats and cultivation and other factors demonstrating a certain trend,in which Shandong and Shanxi Danshen have higher tanshinones content.The average content of Tanshinone IIA is 1.1439mg/g and 0.9794mg/g respectively.The contents of salvianolic acids of Sichuan Danshen are high,and the average content of Salvianolic acid B is 42.7065 mg/g.The content of two components in Shaanxi Danshen showed relatively high overall,and the average contents of Salvianolic acid B and Tanshinone II A achieving 45.6678 mg/g and1.4717mg/g respectively.The situation indicated consistency with general understanding of the current market.3.The composition of different grades of Salvia miltiorrhiza demonstrates a certain trend as well.Taking Sichuan Danshen for an example,from special to third grade,the average content of Salvianolic acid B decreases to 41.6710 mg/g,38.4399 mg/g,36.8293 mg/g and 36.2762 mg respectively.However,the average content of Tanshinone IIA is increased to 0.3720 mg/g,0.3547 mg/g,0.4439 mg/g and 0.5637mg/g.Moreover,the average polysaccharide content is incrementally into 3.35%,3.57%,3.7%,and 3.92% respectively.4.Different drying methods also impact on the two types of ingredients.The contents of salvianolic acid and tanshinones of Salvia miltiorrhizain drying in the sun are higher than that of sweating method.The average contents of Salvianolic acid B and Tanshinone IIA are 62.0814 mg/g and 1.7249 mg/g respectively.5.Identification of several major molds from moldy Salvia miltiorrhiza,which contains Eurotium,Chrysosporium,Aspergillus,Wallemia and Penicillium.Conclusion:1.By analyzing the correlation between traits and intrinsic quality of Salvia miltiorrhiza,it has been illustrated that the content of salvianolic acids showing positively proportional to the diameter of herbs.However,the content of tanshinones moves inversely proportional to the diameter.The content of polysaccharides in Salvia miltiorrhiza demonstrates weakly inversely proportional to the diameter.This is mainly related to the tanshinones distributing in Danshen cutaneous region,and the phenolic acids distributing in xylem.However,these changes with diameter are relatively small compared to the amount contained,thus the impact on clinical medication behaves not significant.At the same time,in order to facilitate market transactions,specification level of Salvia miltiorrhiza has divided due to diameter with rationality.If it is used for the preparation of ingredients such as formula preparations,aiming at one or several indicative components,it can be used to classify each level according to above rules.2.The contents of tanshinones and salvianolic acids from different origins of Danshen are different.Through their external traits,they can reflect the intrinsic quality to some extent,whereas it is hard to make accurate identification lying on appearance alone.It is widely acknowledged that the quality of Danshen in Sichuan and Shandong is good,the two production areas have long history and the grades showing detailed division.Therefore,Sichuan and Shandong Danshen are separately listed as two specifications,and other producing regions are listed as one specification.3.The average content of water-solubility and fat-solubility constituent drying in the sun which is high,meanwhile this method looks simple and costs low.It has become the main processing means at the moment,thus recommended to use sun drying method as the drying method of Salvia miltiorrhiza.4.The composition of Danshen before and after mildew changed obviously.Due to complex components,further analysis is needed in the future.There is less relevant research on the influence to mildew on the quality of medical materials at the moment.It is suggested to enhance the research in this area.
