Objective:1.By comparing the learning-memory scores and neurobehavioral scores of each group of VD rats before and after treatment to verify the effect of warming yang and invigorating the kidney moxibustion in improving cognitive function.2.By observing the infarction and infarct size in rat brain tissue after TTC staining method to obtain the direct evidence that warming yang and invigorating the kidney moxibustion can improve cerebral ischemia.3.By observing the the histopathological changes of rat brain tissue after HE staining to provide the pathological basis for studying the effect of warming yang and invigorating the kidney moxibustion in promoting neuromuscular repair.4.By detecting the expression level of BDNF,NGF,GFAP,Notch1,Hes3 RNA and protein in in hippocampal CA1 region of each group after treatment to obtain the evidence of warming yang and invigorating the kidney moxibustion to Promote the Proliferation of Astrocytes and Generating Nerve Growth Factor and to confirmed its effect in neurons to protect,nutrition,repair and regeneration.To elucidate the effect of warming yang and invigorating the kidney moxibustion on Notch signaling pathway in vascular dementia and to find out the basis of moxibustion to promote the mechanism of nerve regeneration.Method:1.VD model was established by occlusion of the bilateral cervical common arteries and reperfusion.2.The successful 60 VD rat models were divided into four groups: moxibustion group(n=15),medication group(n=15),model group(n=15)and sham operation group as comparison(n=15).Moxibustion group was given warm moxibuste “Dazhui”,“Mingmen” and “Guanyuan”,15 minutes every time and once a day for 4 courses(6 days constitute a therapeuticcourse,with 1 day break).The medication group were treated by lavage of Nimodipine,the course of treatment was same as that of moxibustion group.The sham operation group and model group were given also grasped without any treatment.3.Rats in each group were examined the learning and memory ability through the Morris water maze tests before modeling,before treatment and after treatment.4.TTC staining method was used to detect the infarction and infarct size in rat brain tissue.5.HE staining was used to detect the histopathological changes of rat brain tissue such as apoptosis and edema.6.RT-qPCR and Western blot were performed to respectively detected the expression level of BDNF,NGF,GFAP,Notch1,Hes3 mRNA and protein in hippocampal CA1 region.Result:After molding,compared with the sham group,the escape latency of the other groups were significantly increased(P <0.05).After treatment,compared with the model group,the escape latency of the treatment group was significantly decreased(P <0.05),the infarct size of rats was reduced and the damage of nerve cells in rats brain tissue were alleviated.The expression level of BDNF,NGF,GFAP,Hes3,Notch-1 mRNA and protein were upregulated significantly(P <0.01,P<0.05).Compared with the western medicine group,the cerebral infarct size in the moxibustion group did not decrease significantly;the degree of brain neuron damage was not significantly reduced;the expression level of BDNF,NGF,GFAP,Hes3,and Notch-1 mRNA was significantly increased.(P<0.01,P<0.05);the expression level of BDNF,NGF,GFAP,Hes3,Notch-1 protein were not significantly enhanced(P>0.05).Conclusion:Warming yang and invigorating the kidney moxibustion can regulate the expression ofneurotrophic factors,improve symptoms and learning and memory ability of VD rats,the mechanism that may be related to the regulation of Notch signaling pathway.
