Objective: To compare the expression levels of GAP-43 protein and GAP-43 m RNA in different repair methods to explore the effect of commonly used nerve repair methods,such as end-to-end anastomosis,nerve cannula nesting and autologous epineurium small gap anastomosis,on peripheral nerve injury in rats and the mechanism of peripheral nerve injury repair.The above aims to provide a certain reference value for clinical work.Methods: Two-month-old male S-D rats weighing approximately 250g-300 g were randomly divided into 3 groups with 30 animals in each group.The three groups were end-to-end anastomosis group(Group A),nerve sheathing group(Group B),and autogenous adventitial small gap bridges group(Group C),respectively.In group A,the left sciatic nerve was dissected and in situ end-to-end anastomosis was performed.In group B,nerve cannula nesting was performed after nerves disconnected,and in group C,anastomosis was performed in the small gap of autogenous epineurium.At the 5,7,14,21,and 28 days postoperatively,the nerve samples were removed and Western Blot technique and RT-PCR were performed to detect the expression of GAP-43 protein and its m RNA in the nerve tissue to compare of GAP-43 expression patterns and differences among three different methods.The data was analyzed by ?2 test.p<0.05 was a statistically significant difference.Results: Compared with the end-to-end anastomosis group,GAP-43 protein and its m RNA expression were significantly increased in the neural tube bridging group and the autogenous small-gap bridging group at certain time points,and the difference was statistically significant(P < 0.05)Conclusion: The nerve regeneration chamber constructed by the nerve tube sheathing and the autogenous small-gap bridging is more conducive to the expression of GAP-43 protein and its m RNA than the end-to-end anastomosis,and has a certain role in promoting the repair of damaged nerves.
