| The animal models of tumor has been widely used to simulate human tumor disease,which has become an important part of scientific research and made great contributions to the development of biomedical and the health of mankind.With the development of biotechnology,the types of tumor animal models are constantly enriched,it is essential for exploring the pathogenesis of human diseases and developing new technology and therapeutic drugs.Tumor animal models can be classified into three types:animal model of spontaneous tumor,animal model of induced tumor and animal model of transplanting tumor.The latter two models are widely used,especially the transplanting tumor animal models that can be assigned into two kinds:homologous transplantation and xenotransplantation.In the xenograft model of tumor,the rejection reaction frequently occurred,so we usually choose the immunodeficient animal as the experimental object.This kind of animal’s immune function is not perfect or lacking,so it has a promising application in the research of oncology and immunology.MUC16 not only expressed in normal epithelial cells to play a role of barrier protection,but also have high expression in some cancer cells,making these cells to escape from being killed by NK cells,which will promote occurrence and metastasis of tumors.Due to the complex structure of MUC16 with huge molecular weight and highly glycosylated,it is a big challenge to study the specific function and regulatory mechanism of MUC16 in cancer development.In this study,several tumor animal models related to MUC16 were established to explore the influence of MUC16 on tumor in vivo.The experimental results showed that overexpression of MUC16-C could promote tumor growth with the model of subcutaneous tumor in nude mice.In a metastatic tumor model,the tumor metastases to the liver and lungs.In addition,subcutaneous tumor model can be successfully established with several cancer cell lines that express endogenous MUC16 in high level.Several animal models in this project have laid a foundation for the further study about biological functions of MUC16,it also provides reference for screening and evaluation of anti-tumor drugs. |
