| OBJECTIVE:To investigate the effect of electroacupuncture(EA)on the expression ofPI3K/Akt/m TOR pathways in rats with vascular dementia by using EA JingPoints and Western drug piracetam to treat PI3K/Akt/m TOR signalingpathways.The expression of PI3K/Akt/m TOR signaling pathway was used tostudy the effect of EA related Jing Points on the rat model of vascular dementia.Methods:This study selected 90 clean SD male rats.Before the experiment,the rats werescreened to reduce the impact of avoidance differences.After 1w of adaptivefeeding,the rats were started;before the experiment,rats were trained first.Remove too slow and sensitive mice.Twelve sham-operated rats weregenerated by computer random number method.The VD model was preparedusing a modified four-vessel occlusion method(4-VO).After 4 days ofmanufacture,eligible Morris water maze tests were again used to screen eligibleVD rats in combination with neuroethology scores.Computer random numbermethod was used to generate 3 groups,electroacupuncture group,westernmedicine group and model control group,with 14 rats respectively.Electroacupuncture group was treated with electroacupuncture at Yongquanand Zhongchong points.Western medicine group was treated with piracetam.Itwas a course of treatment for 10 days.There were 3 courses of treatment andthere was a 1 day rest period between treatments.After 2 courses of treatment,the water maze scores and neurobehavioral scores of each group weremeasured.The hippocampal HE staining was performed to observe themorphological changes in hippocampal CA1 region.Immunohistochemistry wasused to detect the expression of PI3 K,Akt and mTOR in the hippocampal CA1 region.Results:1.The neuroethology scores of the model rats were greater than 1 on the 4thday after the model was replicated.The mean escape latency and t(t’)testdifference between the model group and the sham-operated rats water mazeexperiment were both significant.Statistical significance(P<0.01)indicatesthat the scores of learning and recall in the VD model rats were significantlylower,and the model success was achieved.2.After three cycles of treatment,the water maze straddles the number ofplatforms,escape latency,and neurobehavioral scores,the difference betweenelectroacupuncture,western medicine,and model control groups wasstatistically significant(P<0.01,P<0.05),the EA group compared with thewestern medicine group also had statistically significant differences(P <0.01,P<0.05).The results showed that electroacupuncture and Western medicine hadgood curative effect in shortening the escape latency,increasing the number ofcrossing platforms,and reducing the neurobehavioral score of VD rats,and theelectroacupuncture group was more effective than the Western medicine group.3.HE staining in hippocampal CA1 area.The sham operation group had almostnormal nerve cell structure.The cytoplasm was stained purple and the nucleiwere clearly visible.Some visible nucleoli were stained blue.In the modelcontrol group,the structure of the brain tissue was loose,neurons weredegenerated,and the nuclear condensation and staining were deep.Somecytoplasms showed vacuolar changes,and some capillaries were alsosurrounded by congestion.In the electroacupuncture group,most of theneurons were relatively intact and lightly stained.Only a few neurons weredegenerated and necrotic.The western medicine group had more degenerationand necrosis than the EA group.4.Immunohistochemistry assay detected the expression levels of PI3 K,Akt andm TOR in hippocampal CA1 region in each group.Compared with shamoperation group,EA group,western medicine group and model control group,the expression levels of PI3 K,Akt and mTOR increased significantly.Statistically significant(P<0.01).Compared with the model control group,the expressionlevels of PI3 K,Akt,and mTOR in the electroacupuncture group and the westernmedicine group were significantly increased(P<0.01),and theelectroacupuncture group was superior to the western medicine group(P<0.01).The results showed that electroacupuncture has a significant role inimproving the expression of PI3 K,Akt and m TOR in hippocampal CA1 region ofVD rats.Conclusion:1.Electroacupuncture related Jing Points have good results in improvingmemory performance and improving neurobehavioral scores in VD rats.2.Electroacupuncture related Jing Points can activate the PI3K/Akt/m TORsignaling pathway,and thus effectively treat VD,and its mechanism may berelated to the regulation of neural cell proliferation,growth,and differentiationby this signaling pathway. |
