Objective 1.To examine the expression of ?-Catenin and proliferative marker Ki-67 in Primary Retroperitoneal Liposarcoma and evaluate their potential clinical relevance and prognostic values.Meanwhile to investigate factors impacting on local recurrence in case of primary retroperitoneal liposarcoma after complete enbloc resection.2.To analyze the effect of wnt/?-Catenin pathway on SW-872.Methods 1.Clinical data about 90 patients with Primary Retroperitoneal Liposarcoma managed with surgery in our hospital from January 2007 to December 2014 were retrospectively analyzed and all patients were followed up.HE staining,indirect immunofluorescence staining and immunohistochemistry were used to evaluate the impact of ?-Catenin and proliferative marker Ki-67 in Primary Retroperitoneal Liposarcoma.Meanwhile 5 fresh tissue samples were coloected during August 2016 to May 2017.Analysis and comparison the expression of ?-Catenin in five Primary Retroperitoneal Liposarcoma cases and control samples by western blot.Univariate analysis and Multivariate analysis of factors with postoperative recurrence were performed.2.Inhibitor and activator of Wnt/p-catenin signaling are used to anlyse the effect of wnt/?-Catenin pathway on the proliferation of SW-872.Results The high expression of ?-Catenin and Ki-67 in Primary Retroperitoneal Liposarcoma samples was detected.The positive rates of(3-Catenin and Ki-67 were 74.44%(67/90)and 80%(72/90)respectively.The high expression of P-Catenin was significantly related to mitosis,tumor differentiation tumor necrosis,and tumor grade(?2=42.723?40.181?26.052?50.130,P<0.01).Similarly,a significant association was observed between high proliferation index of Ki-67 and tumor size,mitosis and tumor grade(?2=6.065?44.787?8.503,P<0.05).A high Ki-67 proliferation index was detected in association with ?-Catenin over expression too(? 2=103.885,P<0.01).Univariate analysis showed modusoperandi of the tumor resection(? 2=6.946,P=0.008),tumor grade(?2=84.076,P<0.01),tumordifferentiation(x2=43.279,P<0.01),tumornecrosis(?2=3 8.773,P<0.01),mitosis(?2=50.111,P<0.01),?-Catenin overexpression(x2=47.146,P<0.01)and high proliferation index of Ki-67(x 2=51.328,P<0.01)were positively correlated with survival.Multivariate analysis showed that tumor grade(RR=1.968 P=0.031),?-Catenin overexpression(RR=1.564 P=0.037)and high proliferation index of Ki-67(RR=2.551 P=0.000)were independent prognostic indicators for Primary Retroperitoneal Liposarcoma.Western blot analyses showed that ?-Catenin was more highly expressed in the tumour parts of Retroperitoneal Liposarcoma than normal control tissue of the same patients.Meanwhile 3-year local recurrence rate was 62.67%.Univariate analysis showed that tumor size(? 2=12.196 P<0.05),tumor grade(? 2=81.250 P<0.05),tumor differentiation(?2=58.377 P<0.05)and ?-Catenin overexpression(x2=34.866 P<0.05)were all risk factors related to postoperative local recurrence in primary retroperitoneal liposarcoma undergoing complete resection.Multivariate analysis showed that tumor size,tumor grade and ?-Catenin overexpression were independent factors affecting recurrence-free time in primary retroperitoneal liposarcoma(P<0.05).Potently activates the Wnt/beta-catenin signaling pathway can enhance the the proliferation of SW-872 through CCK method and western bloting.Conclusions Overexpression of ?-Catenin protein and a high proliferation index of Ki-67 strongly correlate with poor clinical outcome and reduced survival in patients having Primary Retroperitoneal Liposarcoma.Combined detection of ?-Catenin and Ki-67 has a prognostic value in patients with Primary Retroperitoneal Liposarcoma.
