Part 1 Expression and clinical significance of IL-22 BP,IL-22,STAT3 in the human gastric cancer tissuesObjective To analyze the correlation among the expression of IL-22BP(IL-22 binding protein),IL-22(Interleukin-22)and STAT3(Signal Transducer and Activator of Transcription-3)in human gastric cancer tissues,and discussed the clinical significance of the above factors in combination with clinicopathological parameters.For attempting to provide a new idea in searching for the potential markers to early diagnosis and accurate treatment of gastric cancer.Methods 37 cases of gastric cancer tissues and their matched 37 cases of normal gastric mucosa which adjacent to proximal margin of the surgical resection and beyond 5cm to the tumor region at any rate were collected from the first affiliated hospital of Guangxi medical university from November 2014 to August 2016.All the tissues were confirmed by pathology and there was no infiltration of gastric cancer cells in the surgical margin.The patients who had received radiotherapy,chemotherapy,other anti-cancer treatments beforeoperation or those who were associated with other diseases(such as diabetes,immune diseases,infectious diseases,gastric stromal tumors,other systemic tumors,etc.)were also excluded.After fixing,embedding,and slicing to all of the tissues,immunohistochemical staining SP method was used to observe the expression of IL-22 BP,IL-22,STAT3 in the different tissues.Then SPSS software(version 17.0)was used to analyze the correlation among the above factors,and to explore the relationship between the expression of the above factors with the clinicopathological parameters such as gender,age,invasive degree,the TNM stage,lymph node metastasis of gastric cancer in the patients.Conclusions There exists the highly expression of IL-22 BP,IL-22 and STAT3 in the gastric cancer tissues,and the expression of the three goals arerelated with the invasion and metastasis of gastric cancer,this results suggest that the three may collaboratively participate in gastric cancer.Part 2 Study on the expression levels and clinical significance of IL-22 in the peripheral blood of patients with gastric cancerObjective To detect the expression of IL-22(Interleukin-22)in peripheral venous blood of patients with gastric cancer,and to explore its clinical significance in combination with the clinical records.Methods A total of 37 fresh peripheral venous blood samples were collected from newly admitted gastric cancer patients in the first affiliated Hospital of Guangxi Medical University from November 2014 to August 2016.The gastric cancer patients corresponding to all the specimens were confirmed as gastric cancer by pathology.Patients with other diseases(such as hypertension,diabetes,immune diseases,infectious diseases,gastric stromal tumors,other systemic tumors,etc.)and those who have received cancer adjuvant treatment such as radiotherapy,chemotherapy before surgery were excluded.At the same time,31 peripheral venous blood samples were collected randomly from 31 healthy volunteers as control.Using the real-time fluorescence quantitative PCR Technology(RT-PCR)and enzyme-linked immunosorbent assay(ELISA)respectively to detect the expression levels of IL-22 m RNA in PBMCs and theconcentration of IL-22 protein in serum of all the peripheral venous blood samples,then combined with the clinical and pathological data of gastric cancer,the relationship between IL-22 and the degree of invasion and progression of gastric cancer was evaluated.Finally,SPSS17.0 software was used to complete the statistical analysis.Results The relative expression of IL-22 m RNA in PBMCs of the patients with gastric cancer was significantly higher than that in the healthy controls(2.29×10-4±2.89×10-4 vs 5.90×10-5±6.28×10-5),comparing the difference between the two groups was statistically significant(P < 0.05).The relative expression of IL-22 m RNA in PBMCs of the patients with gastric cancer was related to the TNM stage,invasion degree and lymph node metastasis(all P<0.05),but irrelated with gender and age(all P > 0.05).Compared with IL-22 content,serum IL-22 content in the peripheral blood of patients with gastric cancer was significantly higher than that of healthy controls(193.41±1.71pg/m L vs 81.95±47.38pg/m L),and the difference was statistically significant(P<0.05).Conclusions The expression of IL-22 in peripheral venous blood of patients with gastric cancer is significantly increased,and it's closely related to the clinicopathological features of gastric cancer such as the depth of invasion,lymph node metastasis,which suggests that IL-22 may be a potential marker for clinical evaluation and monitor of gastric cancer.
