Objective:To reveal the expression of Slit/Robo family members and MMPs in the synovial membrane of knee(osteoarthritis OA)and analyze their significance on the pathological changes of synovial membrane in knee osteoarthritis.Methods:19 clinical cases were enrolled in this study.Among these cases,14 were used as the OA group and 5 were used as normal control group.All of the OA group cases had been diagnosed as osteoarthritis with kellgren-lawrence grade III or IV,requiring joint replacement.All cases in the normal control group were of young patients with meniscus injury(corresponding to the Kellgren-Lawrence diagnostic criteria of grade 0)and with onset of the injury within 1 month and without Synovitis showed on MRI.The synovial tissue was derived from the suprapatellar bursa of the knee.The expression of Slit/Robo family members and MMP1-3and MMP8-13 were detected by RNA extraction,reverse transcription,and Real-time PCR.Statistical analysis was relied on Microsoft Office Excel 2016 software and Graph pad Prism v7 biological statistics software and set the significance level ?=0.05.Results:(1)There was no significant difference in the expression of Slitl,Slit2,Slit3,Robo2,Robo4,MMP1,MMP2,MMP3,MMP8,MMP9,MMP10,MMP11and MMP13 between the two groups(p?0.05);(2)The expression of Robot in OA group increased significantly.The expression of the average value of Robol in the normal control group was 1.212±0.3102,compared with 3.042±0.4456 in the OA group.There was a statistically significant difference in the expression of Robo1 between the two groups,with p<0.05(p=0.0313);(3)The expression of Robo3 in OA decreased significantly.The mean expression value was 2.185±0.7007 in the normal control group and 0.5957±0.1176 in the OA group.There was a significant difference in the expression of Robo3 between these two groups,with p<0.05(p=0.0023).Conclusions:(1)The expression of MMP1,MMP2,MMP3,MMP8,MMP9,MMP10,MMP11,MMP13 and Slitl,Slit2,Slit3,Robo2 and Robo4 in synovial tissue of knee osteoarthritis are the same as healthy knee synovial tissue,suggesting that they have no special impact on the pathological changes of synovial tissue of osteoarthritis;(2)Robol is significantly increased in the synovial of knee osteoarthritis,indicating that Robol may promote the proliferation of synovial membrane in knee osteoarthritis while inhibiting inflammation;(3)Robo3 is significantly decreased in the synovial tissue of knee osteoarthritis,indicating that Robo3 may be unassociated with the invasiveness of synovial membrane in knee osteoarthritis.
