| ObjectiveTo study the absorption and metabolism of octopus protein calcium(OP-Ca)in mice and its calcium supplementation effect.It provides the scientific basis for the application of octopus protein calcium as a calcium supplement.MethodsDetermination of chelating rate in octopus protein calcium:The calcium ion content of total calcium and chelated calcium was determined by microwave digestion-flame atomic spectrometry,and the chelation rate of the octopus protein calcium sample was calculated.Animal experiment:Sixty ICR mice were randomly divided into 5 groups:normal control group,low calcium model group,calcium carbonate group and low-and high-dose OP-Ca groups.Except the control group,the other 4 groups were fed with low-calcium diet.Low-and high-dose OP-Ca groups were gavaged with OP-Ca 39.9 and 119.7 mg/(kg·d)once per day,and calcium carbonate group was gavaged with calcium carbonate 119.7 mg/(kg·d).Mice in control group and low calcium model group were gavaged with the same volume of deionized water.Calcium absorption and metabolism test:After 25-day intragastric administration,the feces and urine were collected for 3 days.Urine calcium and fecal calcium levels were measured to estimate absorption rate and retention rate of calcium.Determination of Serum biochemical and femoral index:After 28-day intragastricadministration,all mice were drawn blood from eyeball.The serum levels of serum calcium,alkaline phosphatase(AKP),and total protein in mice were measured using the appropriate kits to investigate the effect of octopus protein peptide calcium on serum biochemical parameters in mice.The mice were sacrificed and the thymus,spleen,and liver of the mice were excised and the thymus,spleen,and liver masses were weighed.The bilateral femurs were isolated and CT scans were performed to obtain CT images of the mice and the cortical bone thickness was measured.The mouse femur density was measured using Archimedes’ principle,and the content of calcium in the femur of the mice was measured by flame atomic absorption spectrometry to investigate the effect of calcium octopine peptide on mouse bone.Result(1)The calcium content of calcium and chelated calcium of octopus protein was 3.78mg/g and 3.25mg/g,respectively,and the chelation rate was 86%.(2)The effect of OP-Ca on calcium apparent absorption rate and retention rate in mice:The apparent absorption rate of calcium in the high and low octopus protein peptide group was 76.30±2.44%,68.27±2.64%and the retention rate was 75.29±2.11%and 66.59±2.34%respectively,which were significantly higher than that in the calcium carbonate group(56.64±2.02%,54.32±1.67%)(p<0.05).(3)The effect of OP-Ca on serum biochemical index in mice:The serum calcium of high-and low-dose OP-Ca groups was 2.23±0.10mmol/L and 2.10±0.08mmol/L,respectively,which was significantly higher than that of the low-calcium model group(1.98±0.10mmol/L)(P<0.05).The high dose OP-Ca groups was higher than calcium carbonate group(2.11±0.13mmol/L)(P<0.05).The serum Alkaline Phosphatase levels in high-and low-dose OP-Ca groups were 12.25±1.73 and 13.10±2.26,respectively,which were significantly lower than those in the low-calcium model group(18.79±3.82)(P<0.05).Compared with calcium carbonate group,the high--dose OP-Ca groups was a significant decrease(P<0.05).(4)The effect of OP-Ca on femur index and femur length in mice:The femur index of high and low dose OP-Ca group were 5.42±0.31mg/g and 5.24±0.12mg/g,respectively,which was significantly higher than that of low calcium model group(4.60±0.33mg/g)(P<0.05).Compared with the calcium carbonate group,the femoral index increased significantly(P<0.05).(5)The effect of OP-Ca on femur density,bone calcium content and cortical bone thickness in mice:The femur density,bone calcium content and cortical bone thickness in the high dose octopus protein group were 1.60±0.07g/cm3,249.12±10.39mg/g,0.70±0.02mm,respectively,which were significantly higher than those in the low calcium model group(1.23±0,06g/cm3,2±7.24±9.30mg/g,0.54±0.02mm).The femur density,bone calcium content and cortical bone thickness in low dose group were 1.44±0.02g/cm3,235.30±8.97mg/g,0.66±0.03mm,respectively,and were significantly higher than those in the low calcium model group(P<0.05).The femoral density,bone calcium content and cortical bone thickness in the high dose group were significantly higher than those in the calcium carbonate group(1.38±0.03g/cm3,230.46±7.90mg/g,0.61±0.01mm)(P<0.05).ConclusionCompared with calcium carbonate,octopus protein-calcium has a higher apparent absorption rate and retention rate.It can maintain the body’s serum calcium and serum alkaline phosphatase levels.It can also increase bone density and increase bone strength to improve the body’s calcium nutritional status. |
