Nano Calcium Kidney Herbs On The Kidney In Rats With Osteoporosis Intestinal Calcium Absorption Mechanism Of Action

Objective: From the aspect of calcium metabolism, exploring the expression of the CaBP-D9K mRNA and protein ,1, 25 (OH) 2D3 receptor mRNA and protein , erkl/erk2 protein in the intestinal mucosa of the osteoporosis rats due to kidney deficiency. To show the episode mechanism of the osteoporosis due to kidney deficiency and explain the effects of the nanometer calcium combined with tonifying kidney Chinese herbs on treating and preventing the osteoporosis. That provided the experimental evidence to the theory of "kidney viscera picture" in TCM.Material and Method: Using the method of ovariectomy, we established the rat model of osteoporosis due to kidney-deficiency; Applying the nanometer calcium and the nanometer calcium combined with tonifying kidney Chinese herbs( big, middleN small dosage) to the rats for 12 weeks, and choosing GUSHUKANG, E2, GAI' ERQI D600 to be the control drugs. Using dual energy X-ray bone density instrument XR-36 model to detect the bone mineral density of the left femur of the rats; We also detected the amount of serum Ca, Mg in the artery, vein, serum TRAP and BGP with the method of biochemistry. Examining the expression of the CaBP-D9K mRNA and protein , VDR mRNA and protein , erkl/erk2 protein in the intestinal mucosa of the osteoporosis rats due to kidney deficiency by the method of RT-PCR and western blotting.Result: (1) The comparation of the experimental animals uterus index: To be compared with the model group, the uterus index of the experimental groups, the nanometer calcium group and E2 group, GUSHUKANG group all increased. And the E2 group increased greatly. But it has no significance difference in statistics. (2) The result of the BMD of the experimental animal femur showed: After 12 weeks, all the BMD of the experimental groups and control groups increased. And the BMD of the nanometer calcium group is similar to the normal group which is the best. The experimental groups' BMD can be obviously increased which is similar to the E2 and GAI' ERQI D600 and is better than the GUSHUKANG group. The BMD of the femoral head indicates: the BMD of the nanometer calcium combined with tonifying kidney Chinese herbs( big, small dosage) groups can be improved which is similar to the normal group. The results of the experimental groups is similar to the control groups and better than the GUSHUKANG group. (3) The symbol of the bone absorption: The results of the serum of the TRAP showed: To be compared with the model group: the serum TRAP of the nanometer calcium combined with tonifying kidney Chinese herbs ( middle dosage) group, E2 group have significant difference in statistics.The other groups have very significant difference in statistics but not achieving the level of the normal group. It has the significant difference to the normal group in statistics. There are no significant difference between all the experimental groups. The results indicated: after the ovariectomy, the content of the serum TRAP of the model group is obviously higher than the other groups; The treating groups and control groups both can lower the TRAP of the OVX rats and have no significant difference in statistics.(4) The symbol of the bone formation: The result of the serum BGP content showed: The BGP of the OVX rats is lower than the normal group. All the drugs can increase the BGP amounts but have no significant difference with the model group in statistics. (5) The results of the serum calcium amount showed: The amount of the serum calcium of the OVX rats is lower than the normal group obviously. All the experimental groups can increase the amount of the serum calcium of the OVX rats and have no significant difference with the normal group in statistics. Meanwhile, all the experimental groups Ca content are better than the control groups. The results of the serum Mg amount showed: The amount of the serum Mg of the OVX rats is lower than the normal group but have no significant difference.
