Purpose:A series of biological cell assays to study the protective effect of ginsenoside CK on cobalt chloride-induced injury in H9c2 cells.The role of ginsenoside CK was confirmed from several aspects including cell injury,oxidative stress,apoptosis and inflammation,investigate the molecular mechanism of action of HIF-1?signaling pathway in ginsenoside CK treatment of cobalt chloride induced injury,provide an experimental basis for explaining the ginseng protection heart function mechanism and product development.Method:Establishment of Myocardial Hypoxic injury model in H9c2 rat cardiomyocytes induced by Cobalt Chloride?CoCl2?;Detection of cell viability using MTT;Kit for detection of lactate dehydrogenase?LDH?,superoxide dismutase?SOD?and malondialdehyde?MDA?;ELISA for Tumor Necrosis Factor?TNF-??and Interleukin-6?IL-6?;Flow cytometry detection of reactive oxygen species?ROS?,apoptosis,calciumion(Ca2+)and mitochondrial membrane potential;Western Blot detection of apoptosis-associated protein,HIF-1?protein and NF-?B p65 protein expression;detection of mitochondrial membrane potential by JC-1 fluorescent staining.Result:1.Compared with control group?Control group?,CoCl2 can significantly reduce cell viability,compared with CoCl2 induction group?Model group?,Ginsenoside CK can significantly increase cell viability,relieves CoCl2-induced cardiomyocyte viability and membrane damage.2.Flow Cytometry results display,compared with control group,CoCl2 significantly increases cellular ROS and Ca2+levels,ginsenoside CK reduces the levels of both,and reduce oxidative stress caused by hypoxia by reducing MDA content and increasing SOD activity.3.Ginsenoside CK can increase Bcl-2/Bax ratio induced by CoCl2-induced injury and decrease mitochondrial membrane potential,so as to reduce the apoptosis-related protein expression to relieve myocardial apoptosis caused by hypoxia.4.Ginsenoside CK reduces inflammatory cytokines TNF-?and IL-6,and the expression of NF-?B p65 protein,reducing the inflammatory response of myocardial cells induced by cobalt chloride injury.5.Ginsenoside CK reduces the increase of HIF-1?protein expression induced by cobalt chloride injury,slow down the damage caused by hypoxia on myocardial cells.Conclusion:Experimental results show that,ginsenoside CK has a good protective effect on cobalt chloride-induced damage to H9c2 cells,the HIF-1?signaling pathway plays an important regulatory role in this process.
