Taxus chinensis var.mairei is a state-level key protected wild plants,is China's unique excellent tree species,widely distributed in our country.Among the people,its seeds and fruit can be used as a medicine;it has good curative effect of kidney disease,diabetes and gastrointestinal disease.Because of the taxol which has stronger antitumor activity extracted from it is widely concerned,but excessive attention cause the excessive use and development of Taxus chinensisvar.mairei resource,even cause a lot of waste of resources,therefore,people began to focus on the parts which is not affect the growth of Taxus chinensisvar.maireiandthe other active ingredients in it gradually.In this paper,we selected the Taxus chinensis var.maireileaves as the research object,optimized the extraction process of polysaccharide and essential oils,and researched the anti-tumor and antioxidant activity respectively.(1)Withmicrowave-assistedsimultaneous distillation and extraction(MA-SDE)method to separate the polysaccharide and essential oils fromTaxus chinensis var.maireileaves.Extraction solvent,particle size,extraction time,microwave power and liquid-solid were selected for single factor experiment,according to the results of the experiment,utilized the response surface software analysis and designed,combined with the actual operation situation,found the optimal extraction conditions:extraction time was 17.5 min,liquid-solid ratio of 22.5(mL/g)and the microwave power of 547 w.the yields obtained for polysaccharides and essential oil under the optimized conditions were 6.39 ±0.12%and 0.27±0.03%,respectively.Compared with the traditional extraction methods,MA-SDEhas obvious advantages of time,its extraction yields are 1.14 times and 1.23 times of the traditional methods,respectively.(2)Taxus chinensis var.maireileaves was studied the activity of polysaccharides in vivo and in vitro antitumor.By the Taxus chinensis var.maireileaves polysaccharide influence of tumor inhibition rate,the body weight and organ on S180 tumor-burdened rat,investigated its antitumor effect in vivo.The research results showed that the Taxus chinensis var.maireileaves polysaccharide has good antitumor ability,the anti-tumor ability of high dose group(600 mg/kg)is strongest,but lower than the positive control group.Using MTT methoddeterminedthat the antitumor effect in vitro of Taxus chinensis var.maireileaves polysaccharide of KBcells,human gastric carcinoma cells(SGC-7901)and human hepatoma cells(SMMC-7721).The experimental results showed that the polysaccharide has a certain degree of cytotoxicityto all subjects cells,and it increased with the increasingconcentrations of polysaccharide.(3)Using the methods of reducing power,DPPH radical and hydroxyl radical scavenging ability of Taxus chinensis var.maireileaves essential oils in vitro antioxidant capacity.The experimental results showed that the Taxus chinensis var.maireilceives essential oil in the three aspects has a certain degree of antioxidant activity,andit increased with the increasing of concentrations of oil,but its antioxidant capacity was lower compared with the V_C control group.
