Taxus chinensis var mairei is unique in China with functions of anticancer.Researches have been reported about the active ingredient of Taxus chinensis place emphasis on barks and leaves,there has little study about the active ingredient in fruit.We optimized the extraction process of polysaccharide in Taxus chinensis fruit and studied it's antioxidation and antitumor activity.(1)With ultrasonic-microwave collaborative assistive technology and through single factor experiment we studied the influence of liquid-solid ratio,extraction time and microwave power on polysaccharide extraction efficiency,then we optimized the technological parameters with Box-Behnken design.The results showed that the optimal extracting conditions were liquid-solid ratio 1:33,extraction time 10 mins and microwave power 650 W.On this condition,the extraction efficiency of polysaccharide in Taxus chinensis fruit can reached 4.33 ±0.15%.This method used the shortest extraction time but had the highest extraction efficiency compared with hot water extraction method,microwave assistant extraction and ultrasonic extraction method.(2)We studied the antioxidant activity of polysaccharide in Taxus chinensis fruit in vitro from three aspects as reducing power,DPPH free radicals elimination and hydroxyl radical scavenging.Results indicated that polysaccharide had different levels of scavenging effect on the three free radicals mentioned aboved,and the reducing power of polysaccharide was lower compared with vitamin C.(3)Using KB cells,human gastric carcinoma cells(SGC-7901)and Human hepatoma cells(SMMC-7721)as the experimental subjects,we explored the antitumor activity of.polysaccharide in vitro,and observed the cells state changes under inverted microscope.The experimental results show,polysaccharide has cytotoxicity to all cells.The effects of polysaccharide on gastric cancer SGC-7901 cells is significantly.IC50 is 2.386 mg/ml.SMMC-7721 cells is not sensitive to the polysaccharide drugs.(4)We further studied the the anti-tumor activity of polysaccharide in Taxus chinensis fruit in vivo.We investigated the influence of the polysaccharide after oral administration on tumour inhibitory rate,body weight change and the relative organ coefficient of S180 tumor-bearing mice.The results showed that the polysaccharide had favourable antitumor activity,and this kind of polysaccharide had the advantages of low toxicity and high efficiency.
