On The Prescription Rule In Treating Myelosuppression With Data Mining

ObjectiveTo explore the rule of Chinese medicine in treating myelosuppression after radiotherapy and chemotherapy,this study mined the valuable information such as the common used Chinese herbal medicine,herb pairs,Chinese medicine groups and the key formulae with the data mining technology including frequency statistics,cluster analysis,association rule and complex network,in order to provide the reference for the clinical treatment of myelosuppression and the development of new drugs.MethodsBy searching the networks such as CNKI,CBM,VIP and Wanfang Data,this study collected and arranged the data of Chinese medicine formulae treating myelosuppression after radiotherapy and chemotherapy.The literatures were put into NoteExpress and the repeated literatures were removed and the database of Chinese medicine formulae treating myelosuppression after radiotherapy and chemotherapy were built.On this basis,the form of the database was transformed into what the computer could recognize,and the names of the herbs in the formulae were standardized by the names recorded in Pharmacopoeia of the People's Republic of China and The Dictionary of Medicinal Plant.R-language,UCINET and NetDraw were used for the frequency statistics,cluster analysis,association rules analysis,complex network analysis and the visualization of results,thereby to summarize the rule of Chinese medicine formulae treating myelosuppression after radiotherapy and chemotherapy.Results 621 formulae for treating myelosuppression after radiotherapy and chemotherapy were collected in this study,including 384 kinds of herbs,in which the high frequency herbs are radix astragali,Rhizoma Atractylodis Macrocephalae,Radix Angelicae Sinensis,Poria,Radix Codonopsis,Radix et Rhizoma Glycyrrhizae,Fructus Ligustri Lucidi,Fructus Lycii,Caulis Spatholobi and Radix Rehmanniae Praeparata.Among all the herbs,the tonifying,warm nature and sweet flavor ones are widely applied,and the main meridian tropisms are liver meridian,kidney meridian,lung meridian and spleen meridian.Association rules analysis showed that 22 herb pairs and groups which are combined by 9 herbs(radix astragali,Rhizoma Atractylodis Macrocephalae,Poria,Radix Angelicae Sinensis,Radix Codonopsis,Radix et Rhizoma Glycyrrhizae,Fructus Ligustri Lucidi,Radix Rehmanniae Praeparata and Caulis Spatholobi),it was also applied to analyze the rule of combination of the high frequency herbs such as radix astragali,Rhizoma Atractylodis Macrocephalae,Radix Angelicae Sinensis,Poria,and Radix Codonopsis.Cluster analysis showed 7 cluster formulae,including 4 formulae which were formed by the modified classical formulae,such as Chinese Angelica Blood-Supplementing Decoction,Four Gentlemen Decoction,Four Substances Decoction and Two Matured Substances Decoction,and the other three are new formulae,which are for supplementing and activating blood,reinforcing healthy qi and dispelling pathogen,and supplementing essence and boosting marrow.Complex network showed four typical formulae which are formed by the modified classical formulae such as Four Substances Decoction,Four Gentlemen Decoction combined with Chinese Angelica Blood-Supplementing Decoction,Radix Adenophorae and Radix Ophiopogonis decoction and Two Matured Substances Decoction,the other four are new formulae,including supplementing qi and blood and also spleen and stomach formula,tonifying kidney and essence and supplementing qi and blood formula,clearing heat and activating blood anticancer formula,and nourishing spleen and stomach and tonifying kidney formula.ConclusionBy using the data mining techniques,the rules of formulae in treating myelosuppression after radiotherapy and chemotherapy shows that the principle of treatment are to reinforce healthy qi,consolidate the root and combined with dispelling pathogen,which focus on supplementing deficiency,it supplements qi and nourishes blood,fortifies the spleen and tonifies kidney,it also combined with blood-invigorating and stasisdissolving,urination-promoting and dampness-percolating,and heatclearing formulae.It treats according to pattern differentiation and it accords to the clinical condition of myelosuppression after chemotherapy,which is healthy qi deficiency with pathogenic qi abundant.
