The Role And Molecular Mechanism Of Linc00673 Targeting Regulation Of ISG15 In Proliferation And Invasive Metastasis In Gastric Cancer

objective: This study aims to elucidate the expression level,clinical significance and possible mechanism of ISG15 in gastric cancer tissues and their cell lines.In addition,the work is to explore and identify that LINC00673 in Linc RNA(long intergenic non-coding RNA)is a potential tumor suppressor.The use of base matching can find that there is a binding site between Linc RNA and ISG15,hoping to provide a valuable experimental basis for targeted therapy by Linc RNA.METHODS: The differences of the expression levels of ISG15 and Linc00673 were detected between normal gastric cells GES-1 and gastric cancer cells BGC823,HGC027,SGC7901 as well as gastric carcinoma and adjacent tissues by means of Western blot and reverse transcriptionpolymerase chain reaction(q RT-PCR)assays.The correlation between ISG15 and clinicopathological features was also ananlyzed.Simultaneously the regulation effect of Linc00673 due to RNAi technology on ISG15’s expression was studied.MTT experiments,scratch tests,Transwell experiments and adhesion experiments were adopted to explore the biological effects of transfected Linc00673.Results:1.The expression of Linc00673 in gastric cancer tissues was significantly lower than that in adjacent tissues,and the similar pattern between gastric cancer cells lines and normal gastric cells lines was showed(P < 0.01).2.The expression of ISG15 in gastric cancer tissues was also apparently higher than that in paracancerous tissues while the expression of ISG15 in gastric cancer cell lines was higher than that in normal gastric cell lines(P < 0.01).The high expression of ISG15 indicated a poor prognosis.3.Silencing the expression of Linc00673 can increase the expression of ISG15 in gastric cancer cell lines.Conversely overexpression of Linc00673 can reduce the expression of ISG15 in gastric cancer cell lines.4.Silencing the expression of Linc00673 and increasing that of ISG15 can inhibit the proliferation,invasion and metastasis of gastric cancer cell lines.However,Overexpression of Linc00673 and decreasing that of ISG15 can enhance the proliferation,invasion and metastasis of gastric cancer cell lines.Conclusion: 1.Linc00673 showed low expression in gastric cancer tissues and gastric cancer cell lines.2.ISG15 showed highly expression in gastric cancer cell lines and gastric cancer tissues.It could be an indicator of the prognosis of gastric cancer.3.Linc00673 can negatively regulate the expression of ISG15 in gastric cancer cell lines and inhibit the proliferation,invasion and metastasis of gastric cancer cell lines.
