Objectives:The effects of intermittent hypoxia on atherosclerosis in rats were studied by establishing the animal model of obstructive sleep apnea-hypopnea syndrome(OSAHS)and the animal model of atherosclerosis.The difference of serum MDA,SOD,sCD-40L,PTX-3 level and pathological changes between rats and normal rats.Methods:Forty-eight 6-week-old Wistar rats of the same cleaning grade were randomly divided into 4 groups.The control group(NC group,n = 12)was fed with.common diet without any other intervention measures,and the atherosclerosis group with as group(AS group,n = 12)was fed with high fat diet to established model of atherosclerosis,OSAHS was simulated by continuous intermittent hypoxia for 9 weeks,continuous intermittent group(CIH group,n=12)were given intermittent hypoxia after the establishment of pharyngeal stenosis.Continuous intermittent hypoxia combined with atherosclerosis group(AS+CIH,n= 12)were fed with high fat diet to establish intermittent hypoxia in pharyngeal cavity stenosis.Arterial blood gas analysis was performed in intermittent hypoxia group,the rats were killed after the experiment,total cholesterol(TC),triglyceride(TG)and serum MDA,SOD,sCD40L,PTX-3 were detected.Myocardial collagen deposition,fibrosis and cardiomyocyte apoptosis were detected by pathological tissue.The degree of atherosclerosis was measured.Results:1.Arterial blood gas analysis:continuous intermittent hypoxia cabin oxygen concentration is the lowest point in rat arterial oxygen pressure:22.6mmHg-32.8mmHg,the lowest oxygen saturation:41.2%-68.6%,when the concentration of oxygen in the cabin is increased to more than 21%,arterial oxygen pressure of arterial oxygen saturation:78.2mmHg-101.7mmHg,95.2%-98.6%.2.The weight of the rats before molding was(193.10 ± 7,80)g.After 9 weeks,the weights of the model group was higher than that before modeling.NC group,AS group,CIH group,AS + CIH group were respectively increased by 14.2%,19.2%,6.9%and 13.9%.There was a significant difference between as AS,CIH group and NC group in heart mass of 9-week rat model.There was significant difference between as AS,CIH,AS+CIH group and NC group in cardiac mass index.(P<0.05)3.The serum TC and TG:there were significant differences between the AS,CIH,AS+CIH group and NC group in 4 week and 9 week.The serum TC and TG were significant differences between 4w and 9w in AS,CIH,AS+CIH group.(P<0.05).4.The serum MDA and SOD:There was a significant difference between group AS,CIH,AS+CIH and group NC at 9 weeks.The serum MDA and SOD were significant differences between 4w and 9w in AS,CIH,AS+CIH group.The increase of MDA and the decrease of SOD obviously in AS+CIH group.5.The serum sCD40L and P-TX-3:there were significant differences between the AS,CIH,AS+CIH group and NC group in 4 week and 9 week.The serum sCD40L and PTX-3 were significant differences between 4w and 9w in AS,CIH,AS+CIH group.The increase of sCD40L and PTX-3 obviously in AS+CIH group.6.Pathology:Apoptotic cells were detected in as AS+CIH group compared with other groups.Sirius red staining showed obvious collagen deposition and fibrosis in rat myocardial tissue.In addition,measurement of intimal thickness of thoracic aorta:there were significant differences between the AS,CIH,AS+CIH group and NC group in 4 week and 9 week.The thickness of thoracic aorta in AS4+CIH group was the most obvious.Conclusions:1.OSAHS may promote and accelerate the thickening of the intima and cause the formation of atherosclerosis.2.OSAHS can promote and accelerate the inflammation,apoptosis,fibrosis of myocardial tissue of CHD,and make ventricular remodeling.3.OSAHS can promote and aggravate the inflammatory response and oxidative stress of CHD.
