At present,chronic kidney disease has been a serious threat to human health,renal interstitial fibrosis eventually developed into end-stage renal disease,and the incidence rate has increased year by year.Traditional Chinese medicine and its effective extracts,Chinese herbal compound have not been obtained in the treatment of renal interstitial fibrosis Take full advantage of traditional Chinese medicine has a good anti-renal interstitial fibrosis application prospects.This study was to investigate the mechanism of the effect of crocin on activation of transcription factor X cassette binding protein 1(XBP1)by activating transcription factor 6(ATF6)in rats with renal interstitial fibrosis.An animal model of renal interstitial fibrosis was established by unilateral ureteral obstruction.Rats were randomly divided into sham operation group,model group,enalapril group,and crocin high-medium low-dose group.Rats were sacrificed on the 14th day after the model was established.Creatinine(Scr)and urea nitrogen(BUN)were measured.The degree of edema in the left kidney of the rat was observed.Renal tubular injury index and renal interstitial fibrosis were evaluated by HE staining.Masson was used.Semiquantitative analysis of renal interstitial collagen area was performed by staining.Apoptosis of renal tubular epithelial cells was detected by TUNEL.The expression of ATF6 and XBP1 protein in each group was detected by Western Blot.The results showed that the scores of Scr and BUN were significantly lower in the treatment group than in the model group at 14 days after surgery(P<0.01).The effects of Scr and BUN were reduced in the high dose group of crocin compared with the enalapril group.Significantly(P<0.05);each treatment group can reduce the kidney weight compared with the model group(P<0.01,P<0.05).The high dose group of crocus can reduce the kidney weight compared with the enalapril group.(P<0.05);Compared with the nelsperidin high-dose group and the enalapril group,there was a difference in the effect of HE staining on renal tubule injury(P<0.05);Compared with the model group,Masson's staining of the relative area of interstitial collagen was significant.Decreased(P<0.01),the degree of renal fibrosis was significantly reduced,there was a significant difference between the high dose group of crotin and the enalapril group(P<0.05);the model group and sham group were found by TUNEL experiment.In contrast,the positive expression of apoptotic cells was significantly different(P<0.01).Compared with enalapril group,the apoptosis index of renal tubular epithelial cells was significantly decreased(P<0.05);Western blotting The expression levels of ATF6 and XBP1 protein were found to be significantly higher in the model group than in the sham group(P<0)..01),compared with the model group,each treatment group was able to effectively reduce the expression of ATF6 and XBP1 protein(P<0.01),and the high dose group of crocus was significantly better than the enalapril positive group(P<0.05).In conclusion,crocin can protect kidney function in rats with renal interstitial fibrosis;relieve hydronephrosis in rats with unilateral ureteral obstruction,improve tubular vacuolization,fibrosis,tubular dilatation,and reduce renal interstitial collagen area The formation of renal tubular epithelial cells decreased apoptosis,decreased protein expression of ATF6 and XBP1 in unfolded protein responses,and inhibited fibrosis formation.
