| ObjectiveTo analyze the difference of expression of cytokines and T lymphocyte subsets in non OA population and knee OA patients with different clinical stages.The Chinese traditional medicine for invigorating kidney and activating blood was carried out for the patients with knee OA,and to explore the changes of the cytokines and T lymphatic subgroups in the patients with knee OA after the intervention of traditional Chinese medicine.Methods40 patients with knee OA were assigned to experimental group(? to ?)by different clinical stages and 10 in non OA volunteers.Extracted the venous blood and knee joint fluid from those persons.The expression level of cytokines was detected by ELISA,and the expression level of T lymphocytes was analyzed by flow cytometry.Meanwhile,the OA groups were treated with Long-Bie capsule(4 times a day,3 times a day,2 weeks).Statistical analysis was made on the changes of indicators in different stages of OA group and non OA group and OA group at different stages before and after taking medicine.ResultsSerum ELISA results:(1)There was no significant difference in the expression of serum IL-1 in each group.(2)IL-6 in stage I was increased,but the difference was not statistically significant compared with that in normal group.While the difference of OA II-IV was statistically significant compared with that of the normal group.After the treatment,IL-6 decreased in all groups,but the difference was not statistically significant.(3)The expression of TNF-alpha in OA patients was significantly higher than that in the normal group,but there was no significant difference in the difference between the stage I and the normal groups,stage II and III gradually increased,stage IV slightly decreased.(4)the serum VEGF in OA patients was significantly higher than that of non OA patients,and the difference in stage I and ? was statistically significant compared with that in stage III and IV.Joint fluid ELISA results showed that:(1)IL-1 beta was a statistically significant difference in normal group,After the treatment the IL-1 beta decreased significantly.(2)IL-6 in OA patients was significantly higher than that in non OA group,and stage II-IV was significantly higher than phase I,but the stage IV was lower than phase III.After the treatment,IL-6 decreased significantly in the stage II-IV.(3)TNF-alpha was significantly higher than that in the non OA group.The stage I-III gradually increased with the OA progression,but stage IV was decreased.After treatment,TNF-alpha in the OA patients was decreased.(4)VEGF in the non-OA patients was significantly lower than that of OA group.With the development of OA,the concentration of VEGF gradually increased.There was no significant change after the treatment,Venous blood cytometry showed that there was no significant difference in the expression of T lymphocyte expression in the whole blood of OA patients in each clinical stage and the normal people.There was no significant difference in those after the treatment.Joint fluid flow cytometry results showed that there was no significant difference in the expression of T lymphocytes in all clinical stages of OA.There was no significant difference in those after the treatment ConclusionThe expression of cytokines in knee OA group was different from that in non OA group,and this difference was more obvious in synovial fluid.The expression level of these cytokines is significantly correlated with the severity of disease.There was no significant difference in the expression of T lymphocyte in the whole blood of the knee OA group than that in the non OA group,but the expression level of T lymphocyte in the knee OA joint was higher than that of the peripheral blood,showing that the local inflammation of the knee was the main manifestation of the knee OA,while systemic inflammatory response may not obvious.Long-Bie capsule can effectively inhibit the expression of inflammatory factors in serum and synovial fluid after intervention,and has no obvious effect on VEGF and T lymphocytes. |
