Objective By using ITRAQ proteomic technology to explore differential protein between luminal subtype of breast cancer and basal-like subtype of breast cancer.we are aiming at looking for targeted protein and gene which can provide a new thinking of the treatment of the basal-like breast cancer.Methods1.The luminal subtype of breast cancer cells MCF-7 cell and basallike subtype of breast cancer cells MDA-MB-231 cells were divided into three groups,each group respectively to extract protein,line proteomic markers related peptides,mass spectrometry analysis,database,then analyses raw letter finally.2.According to the literature,pick out the most experimental significance of 30 protein,line q PCR validated at the transcriptional level,combined with the literature analysis,and then pick out the matching q PCR results from mass spectrometry was statistically significant differences in protein.Results1.Using ITRAQ quantitative proteomic methods identify differences in protein and found that all differences in protein A total of 2796,in which A/B(on behalf of the MDA-MB-231 cells,B represent the MCF-7 cell)raised the differentially expressed proteins has 1591,down 1205 differentially expressed proteins.2.According to the different bioinformatics analysis found that the protein is mainly involved in the process of multicellular organisms,cell adhesion,cell movement,and the main way of participating in metabolic pathways,tumor,etc.,metabolic pathway among the common for glycolysis/dysplasia sugar metabolic pathways,carbon metabolic pathways,glutathione metabolic pathways,etc.Combined with the literature,found that luminal subtype of breast cancer,basal hominins-like subtypes of breast cancer prognosis,the ability of invasion and metastasis increases Jie Ti Xing,comprehensive laboratory technique level at present,looking for feasibility of proto-oncogenes relatively large,so we have to look for basal hominins-like subtype compared with luminal subtypes of breast cancer increases of gene and protein,for the purpose of selected 30 rise in a novel protein of the gene,such as,HELS PLEC-1-22 d,application of q PCR have differences of protein transcription level of verification,3.Pick out mass spectrometry and q PCR consistent protein and the protein expression differences of the three largest glutathione S transferase P,myosin light chain,calcium regulatory proteins-3 were analyzed,and its corresponding related genes for GSTP1,MYL9,CNN3.Conclusion1.The luminal subtypes and basal hominins-like subtypes of breast cancer cells protein differences mainly concentrated in the tumor and related metabolic pathways,these metabolic pathways include glycolysis/sugar dysplasia,GSH metabolism and carbon metabolism,etc.2.17 genetic changes in the experiment and experiment results of mass spectrum,of which the most significant differences in protein change of three proteins for glutathione S transferase P,myosin light chain,calcium regulatory proteins-3,it is possible that can differentiate between the two types of luminal breast cancer subtypes and basal hominins-like subtypes of the important targets.
