Objectives This study intends to prepare of sodium fluoride dopped-10% strontium brushite bone cement,detecte of its physical and chemical properties,and explore its effect on proliferation,alkaline phosphatase(ALP)activity and osteoprotegerin protein expression of mouse pre-osteoblast MC3T3-E1 cells.To prove that incorporation of dopped-10% strontium brushite into sodium fluoride body to promote the role of bone formation,to further improve the material properties,provide experimental basis for clinical bone defect repair.Methods 1 Preparation of brushite cement material by chemical curing method,sodium fluoride was added to brushite in 0.5 wt%,1.0 wt%,and 1.5 wt% to prepare different content of sodium fluoride brushite.The X-ray diffraction experiments,scanning electron microscopy,energy spectrum analysis,and compressive strength of the four materials were used to examine their physical and chemical properties;The use of extracts to detect the proliferation and alkaline phosphatase activity of mouse pre-osteoblast MC3T3-E1.Experimental group: control group,brushite group,0.5 wt % sodium fluoride brushite group,1.0wt% sodium fluoride brushite group,1.5wt% sodium fluoride brushite group,The experimental groups were all extracts of bone cement containing 2% fetal bovine serum.The activity of the cells in each group were measured on the 1,3,5,and 7 days,and the activity of alkaline phosphatase on the third day.2 The same method was used to prepare dopped-10% strontium brushite,doped with 0.5wt%,1.0%wt,1.5wt% sodium fluoride and 10% strontium brushite.Testing the physical and chemical properties of 4 groups of bone cement materials,and the effects of five different cement extracts on proliferation,alkaline phosphatase activity,and osteoprotegerin expression of mouse pre-osteoblasts MC3T3-E1 were investigated.Experimental group: 1.5wt% sodium fluoride brushite group,dopped-10% strontium brushite group,0.5wt% sodium fluoride dopped-10% strontium brushite group,1.0wt% sodium fluoride dopped-10% strontium brushite group,1.5wt% sodium fluoride dopped-10% strontium brushite group.Results 1 The results of XRD and EDX showed that sodium fluoride was successfully introduced into the brushite cement,and the addition of fluoride ions had an effect on the peak of diffraction of the brushite;Scanning electron microscopy(SEM)results showed that the observations of the four groups of cement were all similar to flaky,layered,irregularly arranged under the microscope;The compressive strength results showed that the addition of fluoride ions reduces the compressive strength of brushite,but there was no statistical difference;With the increase of sodium fluoride content,it also promoted cell proliferation and alkaline phosphatase activity,and the promotion of 1.5wt% sodium fluoride was obvious.2 The combination of fluorene and thorium in the brushite with respect to brushite diffraction peaks,had an effect on the surface morphology under scanning electron microscopy.The combination of the two elements was better than the effect of the cement with fluorine alone on cell proliferation and ALP activity,and contains 1.5wt% sodium fluoride dopped-10% strontium for cell proliferation,alkaline phosphatase.And with 1.0wt% sodium fluoride dopped-10% strontium brushite on OPG protein levels in mouse pre-osteoblasts was significantly higher than other groups.Conclusions 1 In this study,we successfully prepared a brushite cement with fluorine and both doped with fluorine and strontium.Simultaneous incorporation of fluorine and strontium reduced compressive strength of brushite;2 Dopped with fluorine brushite could promote the proliferation and alkaline phosphatase activity of mouse preosteoblasts.The best effect was to use 1.5wt% sodium fluoride brushite;3 The addition of 1.5wt% sodium fluoride and dopped-10% strontium could promote the proliferation and ALP activity of preosteoblasts in mouse,and the effect was better than that of dopped with sodium fluorite alone and 10% strontium alone;4 Dopped with 1.0wt% sodium fluoride and 10% strontium brushite promotes OPG protein expression in mouse preosteoblasts.
