| Objective Previous studies have shown that cord blood stem cell transplantation can significantly improve neurological function after middle cerebral artery occlusion in rats.The study of intracerebral hemorrhage has been rarely reported.In order to further explore the effect of transplanted stem cells and the mechanism of promoting the repair,we investigated the effect of human umbilical cord blood mononuclear cells(HUCB-MNCs)in intracerebral hemorrhage rats by microPET-CT、Longa scores and RT-PCR.Method We made rat intracerebral hemorrhage model by autologous blood injection.The human cord blood mononuclear cells isolated from human umbilical cord blood were transplanted into the hematoma around in terms of 1 × 10~7/ml after 24 hours of successful model construction.Control group is not receive any treatment.The changes of metabolic changes and neurological function scores of brain injury in rats were evaluated by 18F-FDG PET and Longa scores before and after transplantation.The expression of neuron-specific markers,MAP2 and NSE,were measured in the brain at 3,7,14 and 21 days after transplantation.Results 1.The transplantation group rats neural function score was lower than the control group,there was significant difference(F= 22.352,p<0.0001);before transplantation and third days after transplantation in two groups of rats neural function score have no significant difference(P>0.05);there was significant difference between the 7,14,21 days of the two groups of rats neural function score after transplantation(P<0.05).2.Glucose metabolism of Hematoma Center(SUV%)was significantly higher than control group,there was significant difference(F=49.69,p<0.001);the control group rat brain glucose metabolism of center did not change with time,the difference was not statistically significant(P=0.553>0.05);sugar metabolism of center in experimental rats changed with time,the difference was statistically significant(P=0.031<0.05);differences of glucose metabolism was not statistically significant(P=0.209>0.05)before transplantation and transplantation after third days in control group and experimental group,the rest of the time,the difference of glucosemetabolism levels of two groups were statistically significant(P<0.05).3.Glucose metabolism of Hematoma around(SUV%)was significantly higher than control group,there was significant difference(F=7.668,p=0.024);sugar metabolism of around in experimental rats changed with time,the difference was statistically significant(P=0.007<0.05);the control group rat brain glucose metabolism of around changed with time,the difference was statistically significant(P=0.018<0.05);in addition to before transplantation and transplantation after third days differences in glucose metabolism was not statistically significant(P=0.209>0.05),the rest of the time,the difference of glucose metabolism levels of two groups were statistically significant(P<0.05).4.The rats at different time points,volume generation gradually decreased,there was significant difference(F=556.539,P=<0.0001);each group are not same trend in each time point,the experimental group volume decreased compared with the control group(F=8.855,P=0.018).5.The experimental group MAP2 and NSE increased in seventh days,and then decreased;MAP2 expression difference between control group and experimental group have no significant changes(F=1.49,p=0.29>0.55);the expression of MAP2 in transplantation group changed with time,the difference was statistically significant(P=0.033<0.05);the control group expression of MAP2 did not change,the difference was not statistically significant(P=1.00>0.05);human derived MAP2 expression in transplantation group did not change with time,have no significant difference(P=0.42>0.05);NSE expression difference between control group and experimental group have no significant changes(F=6.09,p=0.069>0.55);NSE expression in transplantation group changed with time,the difference was statistically significant(P=0.007<0.001);the expression of NSE in control group did not change with time,have no significant difference(P=1.00>0.05).Conclusion This study shows that transplantation of umbilical cord blood stem cells can promote intracerebral hemorrhage rats function recovery and the therapeutic mechanism may be related to the promotion of endogenous nerve regeneration. |
