Objective: To demystify the expression of mi R-3619-5p in prostate cancer.(PCa)tissues and Pca cell lines.To explore its role of inhibition of cell cycle and molecular mechanism of mi R-3619-5p associated with PCa cell lines.Methods: To investigate the expression of mi R-3619-5p we used q PCR in several prostate cancer cells and a normal prostate epithelium cell line,and perform a FISH with some clinical prostate biopsy specimens.The biological significance of mi R-3619-5p in prostate cancer cells was assessed by a series of in vitro assays(MTS,colony formation assay).Results:We identified a new endogenous mi R-3619-5p which was decreased in prostate cancer tissues and cells compared to corresponding normal controls.Over-expression of mi R-3619-5p readily induced p21 gene expression by directly targeting the putative site in promoter.Besides,mi R-3619-5p possessed considerable capacity to inhibit prostate cancer cells DU145 and PC3 growth,and downregulate several p21 downstream genes,such as Cyclin D1,CDK4 and CDK6.Conclusions:These findings provide insight into that mi R-3619-5p,which was decreased in prostate cancer tissues and cells,acted as a tumor suppressor by targeting p21.
