Background and Objective:MiR-7-5p and mi R-451 are important members of the mi RNA family,which have been proved to be significantly down-regulated in various human tumors and act as tumor inhibitors in the occurrence and development of tumors.However,little is know about their role in endocrine malignancies.The aim of this study is to detect the expression levels of mi R-7-5p and mi R-451 in formalin fixed paraffin embedded(FFPE)tissues of papillary thyroid carcinoma(PTC)patients and analyze their potential clinical significance.Methods:Real-time fluorescence quantitative polymerase chain reaction(q RT-PCR)was employed to detect mi R-7-5p and mi R-451 expression levels in 101 patients with PTC and 40 patients with nodular goiter(control group)in FFPE tissues.The relationship between mi R-7-5p and mi R-451 expression levels and clinical pathological features of PTC patients,such as gender,age,tumor size,whether or not lymph node metastasis,tumor multifocal,tumor stage and capsular invasion situation,was further analyzed.All statistical analysis was used SPSS 20.0 software package.Results:(1)q RT-PCR results showed that the expression levels of mi R-7-5p and mi R-451 in PTC patients were significantly lower than those in the control group,both of which were statistically significant(all P<0.001).(2)Spearman correlation analysis showed that mi R-7-5p and mi R-451 expression levels had a high correlation in patients with PTC(r=0.537,P<0.001).The lower mi R-7-5p expression level,the lower mi R-451 expression level was,vice versa.(3)ROC curve analysis showed that difference expression of mi R-7-5p and mi R-451 in PTC patients and the control group could effectively identify the PTC and the control group.mi R-7-5p's area under curve(AUC)was 0.895,with sensitivity of 80.2% and 82.5% of the specific degree at cutoff value of 0.0712;The AUC of mi R-451 was 0.768,and its sensitivity and specificity were 62.4% and 82.5%,respectively,at cutoff value of 0.0507.(4)The relationship between the expression level of mi R-7-5p and mi R-451 and the clinicopathological characteristics of PTC: in the PTC,the expression level of mi R-7-5p in tumor with larger diameter group was significantly lower than the tumor with smaller diameter group(P<0.05);and the low expression of mi R-7-5p was related to the advanced stage of tumor in PTC patients,III/IV PTC patients showed significantly lower mi R-7-5p expression than I/II PTC patients(P<0.05).The expression of mi R-7-5p had no relationship with the patient's gender,age,lymph node metastasis,capsule invasion and multifocality(all P>0.05).There were no statistically significant difference between the expression of mi R-451 and the clinicopathologic features of the patients in this study(all P>0.05).(5)According to univariate/multivariate logistic regression analysis,mi R-7-5p was significantly correlated with the occurrence of papillary thyroid carcinoma(P<0.001).Conclusions:1.The expression levels of mi R-7-5p and mi R-451 in PTC tissues were significantly lower than control;2.Mi R-7-5p and mi R-451 could be used as potential biomarkers for the diagnosis and verification of PTC patients;3.Mi R-7-5p was associated with some aggressive characteristics of PTC;4.Mi R-7-5p may be an independent risk factor for PTC.
