Objective: To observe the protein expression levels of PER3 and Notch1 in normal colorectal mucosa,colorectal adenoma and colorectal adenocarcinoma,explore the relationship between the occurrence and development of colorectal adenocarcinoma and their clinicopathological features.Methods:To collecte 10 cases of normal colorectal mucosa,25 cases of colorectal adenoma and 50 cases of colorectal adenocarcinoma.The expression of PER3 and Notch1 protein in the above three tissues are detected by immunohistochemistry.The sum of intensity and number of positive cells in the same type of cells is used to jugement the protein expression levels of PER3 and Notch1.To investigate the expression of these two indicators and their relationship with clinicopathological features,and analysis both correlation in colorectal adenocarcinoma.Results: PER3 and Notch1 are expressed in colorectal adenocarcinoma.The positive rates of PER3 in normal colorectal mucosa,colorectal adenoma and colorectal adenocarcinoma are100%(10/10)?80%(20/25)and 56%(28/50),compared with adenocarcinoma group,there is significant difference between adenoma group and normal group(P <0.05),there is no significant difference between adenoma group and normal group(P <0.05);The positive rate of Notch1 in the above tissues are 10%(1/10),52%(13/25),and 74%(37/50),compared with adenocarcinoma group,there is significant difference between adenoma group and normal group(P <0.05),and there is no significant difference between adenoma group and normal group(P> 0.05).The expression of PER3?Notch1 in colorectal adenocarcinoma are associated with lymph node metastasis,tumor clinical stage,differentiation and depth of tumor invasion(P <0.05),but there are no correlation with age,gender,tumor location and tumor size(P>0.05).Compared with lymph node metastasis group,the positive expression rate of PER3 is less than no lymph node metastasis group(P <0.05),and the positive expression of PER3 is related to the stage and differentiation of tumors.The higher tumor stage,the lower degree of differentiation and the positive expression rate of PER3 is down-regulated(P < 0.05),but Notch1 is opposite.There is a negative correlation between PER3 and Notch1(r =-0.438,P<0.01).Conclusion:1.Compared with normal tissues and adenoma tissues,the positive expression rate of PER3 in adenocarcinomas is gradually decrease,while Notch1 is opposite,indicating that abnormal expression of PER3 and Notch1 are associate with the occurrence and development of colorectal cancer.2.In colorectal adenocarcinoma,PER3 and Notch1 are associate with lymph node metastasis,tumor clinical stage,differentiation degree and depth of tumor invasion.,indicating that decreased expression of PER3 and elevated expression of Notch1 may affect the transfer ability and malignant degree of colorectal adenocarcinoma.The shows PRE3 and Notch1 may be used as new indicators for prognosis of colorectal cancer in the future.3.The expression of PER3 in colorectal adenocarcinoma tissues is negatively correlated with that of Nocth1.
