A large number of studies have confirmed that Cordyceps polysaccharide has anti-tumor,anti-oxidation,immune regulation,liver protection,kidney protection and other pharmacological effects.Fermented cordyceps powder?Cs-4?that Chinese scientists isolated from natural Cordyceps sinensis to obtain Cs-4 Strain,which was artificially fermented and cultured.This experiment used fermented Cordyceps powder as material.The crude polysaccharide is obtained by water extraction,alcohol precipitation,reconstitution,dialysis,concentration and freeze-Drying.The crude polysaccharide was isolated and purified by DEAE-cellulose ion exchange column and sephadex-G-100 column chromatography.The content of sugar was measured by phenol-sulfuric acid method,the relative molecular weight was measured by high-performance gel permeation chromatography,and the monosaccharide was determined by capillary electrophoresis.The mouse splenocytes were used as target cells to investigate the effect of crude polysaccharide on splenocyte proliferation and secretion of IFN-? in vitro.It was found that the proliferation activity of splenic cells in the abdominal cavity can be enhanced and a tendency to increase the secretion level of cytokine IFN-?;Using mouse peritoneal macrophages as target cells to investigate the effects of crude polysaccharide on the secretion of TNF-a by giant cells in vitro.The results found that the secretion of TNF-a from cytokines can be significantly increased;L-O2 cells and The model of high concentrations of Con A induced L-O2 cell injury as the research object,Exploring the effect of polysaccharide components and polysaccharide constituent on the proliferation of L-O2 cells and that damage.The results showed that polysaccharide components and polysaccharide constituent can promote the proliferation of L-O2 cells,and high concentrations of Con A can effectively inhibit the proliferation of L-O2 cells.After addition of polysaccharide components andpolysaccharide constituent,this inhibition is greatly reduced,and it Shows the dose-effect relationship in a certain range.In a word,fermented Cordyceps sinensis polysaccharides can exert immunoregulatory effects by affecting peritoneal macrophages and spleen lymphocyte-s,but Immune regulation and its mechanisms need further study.The fermented Cordyceps sinensis polysaccharides can promote the proliferation of L-O2 cells and protect the L-O2 cells that damaged by Con A.The mechanism of proliferative and protective effects needs further study.
