| In order to efficiently use and develop the medicine edible food which had healthcare function and medical value to prevent and cure diabetes. Based on Cordyceps sinensis(Berk)Sacc screen the optimizing liquid medium, improving the production rate of effective ingredients, and extracting polysaccharides of Cordyceps sinensis(PCS). Acute toxicity test verified the safety. To study the anti-hyperglycemic effect of the combination of PCS and Chinese herb on STZ diabetes mice, screening the best PCS combination. Extracted of Chinese herb hypoglycemic active ingredient and combined with the PCS, screening the best PCS complex and dosage. Established glucose predicts system, querying and predicting hypoglycemic indicators of diabetic mice. The results are as follows:(1)The optimization of cordyceps mycelium cultivationAdded antioxidants KI, seabuckthorn flavonoids, grape seed extract in the cordyceps sinensis fermentation culture medium to determination of index. Three kinds of antioxidants were beneficial to the cultivation of cordyceps sinensis and the generation of active ingredients. KI group of mycelium was the highest biomass and PCS yield, safe non-toxic. It could be used as the optimal antioxidants. In KI=0~5 g/L, cordyceps mycelium dry weight, the content of PCS, PCS antioxidant activity increased with the increase of KI concentration increasing. KI=5 g/L, it produced the largest plant biomass, PCS yield, and the strongest inoxidizability of PCS. So KI=5 g/L was best concentration.(2)Screening the hypoglycemic effect of 6 kinds of cordyceps polysaccharide combinationTo choose PCS combination 6 kinds of Chinese herbal medicine hypoglycemic indicators of STZ diabetes mice, screening the optimal combination. STZ diabetic mice model was established, screening the optimal concentration of PCS(200 mg/kg). PCS(200 mg/kg)combined with 6 kinds of Chinese herbal(150 mg/kg)PCS combination, found that 6 kinds of PCS combination of STZ diabetes mice was strong hypoglycemic effect, was better than individual taking the effects of PCS. Best comprehensive index was PCS and physalis group. For STZ diabetic mice, has the best obvious role in lowering blood glucose, improve body characteristics, strengthen the glucose tolerance, reduce the alpha-glycosidase enzymes, at the same time can obviously rises high oxide dismutase(SOD)activity, lower malondialdehyde(MDA)content, has good antioxidant activity.(3)The hypoglycemic effect of cordyceps polysaccharide complexTo further study of PCS and physalis combination, screening the optimal PCS complex. By comparing the hypoglycemic effect of compatibility of PCS with Polysaccharide from Physalis alkekengi L. var. franchetii Makino( PAVF), and Polysaccharide from fruit calyx of Physalis alkekengi L. var. franchetii Makino(PPSB), selected the optimal complex─PCS and PAVF complex, screened the optimal dose. It was found that the longer the term of PCS and PAVF complex, the greater the dose, the more obvious hypoglycemic effect. Selected the optimal dose for PCS and PAVF complex was 200 mg/kg+175 mg/kg. It could effective hypoglycemic, alleviate symptoms of “a little more than three diseaseâ€, and improve the sugar tolerance ability, inhibition of alpha-glycosidase enzymes. PCS and PAVF complex is expected to become effective hypoglycemic food ingredient.(4)The establishment of the glucose predicts systemGlucose predicts system was software to query and predict hypoglycemic indicators of diabetic mice. User interface was divided into two modules, the first module to blood sugar query system, the second module prediction system for blood sugar.
