Objective:To study and compare the effiency of micro plasma radio frequency versus intense pulse light(IPL)for the treatment of pigmentation induced by ultraviolet light in guinea pigs and the effect on c-kit and PAR-2expression of the skin in the skin were compared.The possible molecular mechanism of the two cases of skin pigmentation was preliminarily explored.Methods:In this paper,Experiments were performed on guinea pig.After depilation,narrow band ultraviolet B(NB-UVB)irradiation was used to induce pigmentation of the skin(parameters:0.9J/cm~2、once a week for 3weeks).After models were completed,the skin of the model area was divided into A、B、C three groups.A group:no treatment;B group:given micro plasma radio frequency technology therapy.Therapeutic process:fixed-point H-TIP、output power:30Watt、time of operation:0.6s、total energy:18J、treat one time;C group:given intense pulsed light therapy.Therapeutic process:smear about 1~2mm thick cold gel before irradiation,then treat one time with these parameters as follows:energy density 8J/cm~2,Choose the treatment mode of AFT 570nm,pulse width 12 ms,facula area 10 mm×30mm,treat one time.Respectively,Biopsies were taken from A、B、C each treatment sites at the time of before and after the model establishment、2weeks and 4 weeks after treatment,in addition took pictures at the same time.Evaluate the variation of skin colour by visual inspection.The skin tissues were dealed with HE staining、melanin granules Masson-Fontana staining、immunohistochemistry staining of c-kit receptor and Protease-activated receptor-2(PAR-2).Finally,The changes of skin color,tissue structure,the Synthesis of melanin granules and the expression of c-kit and PAR-2 protein were evaluated in each group.Results:1.The change of skin color with naked eyeAfter UVB,The skin color of guinea pig irradiated area was obviously darkened.Pigment spots were dark brown and evenly distributed.2 or 4 weeks after treatment,the skin color decreased slightly in group A.The skin color faded obviously in both B、C treatment groups.2.The integrality of skin tissue structure and qualitative evaluation of melanin granulesHE staining showed that the epidermal structure and tissue of each group of guinea pigs were intact.There was no obvious disorder in the arrangement of epidermal cells too.The distribution of melanin granules in epidermis was increased significantly after UVB irradiation.After treatment,the distribution and density of epidermal melanin granules in group B and group C was significantly lower than that in group A.3.Statistical Analysis of the content of melanin granulesThe content of melanin particles in the epidermis after NB-UVB irradiation increased significantly compared with that before irradiation and the difference was statistically significant(P<0.05).2 and 4 weeks after treatment:Melanin granules in group B、C were lower than those in group A in the same period(P<0.05).Melanin granules in group C were lower than those in group B(P<0.05).The content of melanin granules was compared at 2 weeks and 4 weeks after treatment,and the difference was statistically significant(P<0.05).There was interaction between time factor and grouping factor.4.Statistical analysis of the protein expression of c-kit and PAR-2The expression intensity and quantity of c-kit、PAR-2 were both increased after ultraviolet irradiation.Differences were significant before and after model establishment(P<0.05).c-kit protein:After 2 weeks of treatment,the expression of c-kit in group C was lower than that in group A and the difference was statistically significant(P<0.05);There was no significant difference in c-kit expression between group B and C(P>0.05).4 weeks:The expression of c-kit in B and C group was significantly lower than that in group A,and the difference was statistically significant(P<0.05).The expression of c-kit in group C was lower than that in group B(P<0.05).PAR-2 protien:2 and 4 weeks after treatment:The expression of PAR-2 in B、C group was lower than that in group A in the same period,and the difference was statistically significant(P<0.05);The expression level of PAR-2 in group C was less than that of group B in the same period,and the difference was statistically significant(P<0.05).Conclusions:The micro plasma radio frequency technique can obviously improve the skin pigmentation induced by narrow band ultraviolet radiation in guinea pigs,and also verify the exact effect of intense pulsed light on the improvement of skin pigmentation.Compared with micro plasma radio frequency technique,intense pulsed light has more advantages in the treatment of skin pigmentation.On the one hand,the mechanism of the treatment of skin pigmentation may be related to the decrease of c-kit receptor,which may lead to the inhibition of melanocyte activity,growth and proliferation.On the other hand,it may be related to the decrease of PAR-2 receptor expression and interfere with the transport process of melanin granules,which may affect the melanin metabolism by interfering with the transport of melanin granules.
