Comparisons Of The Interferon Alpha-2a And Interferon Alpha-2b On Immunogenicity And The Immunotoxicity

Objective:To investigate the immunogenicity and immunotoxicity in Kunming mice after intraperitoneal injection of interferon alpha-2a(IFN ?-2a)and interferon alpha-2b(IFN ?-2b)Methods: Kuming mice were randomly divided into positive control group(cyclophosphamide,group P)?IFN ?-2b high dose group(group A)and low dose group(group B)?IFN ?-2a high dose group(groupC)?low dose group(groupD)and negative control group(solvent control,groupN),intraperitoneal injection for 6 weeks and rat tail vein blood collection once a week.The immunogenicity was compared by the levels of serum IFN? and serum IFN? antibody that were detected by ELISA and and the renal immune complexes that were detected by C3 immunohistochemical staining.The body weight,organ index,hematologic traits(WBC?PLT?RBC?HGB?MCV?MCH?MCHC?HCT?LY?GR?LY%?GR%),the pathological sections of spleen and immunoglobulin(IgA,IgG,IgE)were used to compare the immunotoxicity of two types of IFN ?.Results :When compared with group A,the serum concentrations of IFN? in group B and C were obviously elevated,the serum concentration of IFN? in group C was also higher than group D(P<0.05=B and D only showed significant differences in the fourth week.The concentrations of serum IFN-AB in group A was higher than B in the 1,3 and 5 weeks;groupC was higher than D in the 1,3 and 5 weeks;group A was higher than C in five weeks;group B was higher than D in the 1?2?3?5weeks(P<0.05).Renal C3 complement immunohistochemistry showed that A and C;B and D compared to immune complex precipitation was different(P < 0.05).The weight gain of group P,A,B and D was different from that of groupN(P <0.05).The results showed that the liver index of groupD was higher than groupN,the spleen index of A,B,C,D were much more higher than that of positive control group P,and B group was significantly higher than N group(P<0.05.The levels of WBC,PLT,MCH,LY #,NE # in each intervention group were higher than that of group N,and were lower than that of group P.(P <0.05)There were no differences in concentrations of IgA and IgM,but when compared to groupP,the levels of IgG in group A,B,C and D were higher(P<0.05).Conclusion:1.There were some differences in the immunogenicity of IFN alpha-2a and IFN alpha-2b,2.There was no significant difference between IFN-2a and IFN alpha in-2b.
