Paliurus ramosissimus?Lour.?Poir,the main chemical composition is triterpenes,total flavonoids and coumarins and so on,the triterpenes compounds had good antitumor activity.The antitumor activities of paliurusene,betulinic acid,messagenic acid B and Triterpenoids against gastric cancer cell lines AGS and HGC-27 were determined by MTT assay and CCK-8 assay respectively.The results showed that the method of CCK-8 was more simple and quick.The results are more stable and reproducible.However,the trend of the inhibition rates obtained by the two methods is consistent,and the IC50 values are similar.The in vitro anti-tumor activity of paliurusene,betulinic acid,messagenic acid B and total triterpenoids on a variety of tumor cells was investigated.The results showed that the above components were all on liver cancer,cervical cancer,lung cancer cells,and gastrointestinal cancer cells.It has a certain anti-tumor activity in vitro.Among them,paliurusene has the strongest anti-proliferative effect,and paliurusene,messagenic acid B and total triterpenoids have stronger inhibitory effects on gastrointestinal tumors in vitro.Comparing the total triterpenoids and betulinic acid,the total triterpenoids had better anti-tumor activity against various tumor cells in vitro.In this study,the shake-flask method was used to determine the oil-water equilibrium time curve of betulinic acid and the total constituents of the triterpenes of paliurusene and the betulinic acid and the partition coefficients of oil and water under different pH conditions.The results showed that the betulinic acid and the triterpenes were almost equal to the balance of oil and water after shaking for 10 h.Betulinic acid and the paliurusene,betulinic acid in the triterpenoids apparent oil-water partition coefficients were 2.913.12?2.913.15?1.962.13,The total triterpenoids can be used for absorption in the gastrointestinal tract.In order to investigate the total triterpene absorption of the seeds,we used 0.5%sodium dodecyl sulfate to increase the solubility of the total triterpenoids.In this study,gastrointestinal absorption was measured in vivo by the method of gastrointestinal absorption,the experimental study of paliurusene and betulinic acid absorption in the stomach and different pH conditions,different quality Concentration and absorption of different parts of the intestinal absorption.Betulinic acid and paliurusene in the stomach were 9.733%and 13.811%,respectively.Betulinic acid and paliurusene had different pH in intestinal absorption,and the absorption rate constants of different concentrations were basically the same 0.112±0.027h-1,0.127±0.007h-1,although the intestinal absorption was slightly different but no significant difference.The intestinal absorption of the components of the total triterpenoids in the concentration range was a passive diffusion mechanism with no specific site of absorption.Using LC-MS method,taking oleanolic acid as internal standard,the concentration of betulinic acid in rat plasma was determined.The method is stable and has good sensitivity and is suitable for the determination of betulinic acid in rats.
