Investigation Of Interleukin-14 Alpha In Patients With Gastrointestinal Non-hodgkin’s Lymphoma That May Result From Autoimmune Disease

Objective We constructed interleukin-14 alpha transgenic mice model to reproduce the occurrence and development of Sjogren syndrome and now we use this mice and clinical study in lymphoma patients to investigate the role of IL-14α plays in autoimmune associated GI-NHL.Method 148 cases of patients with lymphoma(including 57 GI-NHL patients)were enrolled in our research,and the serum samples from 90 cases normal control and pathological sections of 135 patients with lymphoma were collected.ELISA was used to detect the expression of autoantibodies in serum of all patients,and all patients were divided into two groups,autoimmune antibody positive(Ab+)lymphoma group and autoimmune antibody negative(Ab-)lymphoma group according to the results.Next,Western Blot and RT-PCR were used to detect the expression of Interleukin-14 alpha(IL-14α)protein and gene in samples.Immunohistochemical staining was conducted to detect the expression of protein in tissue sections of 6 patients with GI-NHL.Detect the expression of IL-14α in lymphoma cells(Raji)to verify the antibody effect.Result The gastrointestinal non-Hodgkin’s lymphoma patients had a higher expression of autoantibodies than that of NGI-NHL patients(p<0.05).And the former was significantly higher in the proportion of positive in patients than the latter.Cytokines IL-4(p<0.05),IL-17(p<0.05)was higher in patients compares to nongastrointestinal lymphoma.Of the 57 cases of GI-NHL patients,IL-14α was positive in 18 patients(31.57%),and in the 91 cases of NGI-NHL,the expression of IL-14α antibody was positive in 15 patients(16.48%).There was significant statistical difference between the two groups(p=0.015).The percentage of IL-14α positive expression in patients with Ab+ lymphoma was 52.5%,higher than that in AbLymphoma Group(16.25%,p<0.05)and normal control(11%,p<0.05)In patients with positive autoantibodies(Ab+ lymphoma),12 out of 25 GI-NHL patients express positive IL-14α(48%)and 26.6% in NGI-NHL patients.While,in patients with negative autoantibodies(Ab-lymphoma),24% of GI-NHL patients express positive IL-14α and 14.47% in NGI-NHL patients.Conclusion The expression level of IL-14 alpha is higher in autoimmune related gastrointestinal lymphoma than non-gastrointestinal lymphoma,thus,IL-14α may be used as a serological diagnosis index for autoimmune diseases associated with nonHodgkin’s lymphoma of the gastrointestinal tract,and early detection can guide clinical treatment and prevent the occurrence of serious complications.
