| Aims Circulation level of adipokine omentin is negatively related with outcomes of ischemic stroke.However,the single nucleotide polymorphism(SNP)of omentin is still unknown.Thus,our study aims to research the correlation between SNP of omentin and acute incidence of ischemic stroke.Methods According to the principle of nested case-control study,we firstly choose the case and control group from a two-year(2013-2015)cohort,which is organized by Xi’an stroke center.Patients with newly onset ischemic stroke are distributed to case group while without any history of stroke ones to control group.Then,we record and analyze the general demographic and clinical information of enrollments.And after the gene extraction from preserved peripheral blood,the promoter and every exon sequence of omentin gene are duplicated with the means of PCR.Later,the SNPs locating in promoter and every exon regions are identified comparing the tested sequence and standard one from NCBI database.Furthermore,we adopt added model to analyze the relationship between detected SNPs and different phenotypes,in which binary ones,such as acute ischemic stroke,blood pressure and so on,are applied with cox regression analysis and continuous ones,such as total cholesterol,homocysteine etc.,with linear regression analysis.Besides,the relation of tested SNPs and acute ischemic stroke between different gender is also explored with stratification analysis.At last,we employ multiple regression analysis to control confounding factors,which includes high blood pressure,diabetes,coronary artery diseases,body mass index(BMI),homocysteine,total cholesterol,total triglyceride,low density lipoprotein,media thickness of carotid artery,as well as plaques of carotid artery,to identify independent correlation between omentin SNPs and acute ischemic stroke.Results 1.Among Shann’xi population of Han ethic group,we detect 21 SNPs in promoter and all exons sequences of omentin,in which rs2274908 is synonymous mutation,while rs2274907 is missense mutation.The 109th amino acid varies from aspartic acid to valine with its corresponding rs2274907 mutants from adenine to thymine.2.Independently from high blood pressure,coronary heart diseases,BMI,homocysteine,total cholesterol,total triglyceride,low density lipoprotein,media thickness of carotid artery,as well as the existence of plaques in carotid artery,when the base of rs12094513 and rs60429964(thymine and guanine respectively)is substituted by cytosine,the possibility of occurrence to acute ischemic stroke would increase 22 times(P>0.05)in males;however,in terms of females its possibility would decrease 185times(P>0.05).3.If rs2274907 mutants from adenine to thymine,the risk of acute ischemic stroke would decrease 1 time(P>0.05).Conclusions 1.In males the mutation of rs12094513(C>T)and rs60429964(C>G)trends to the occurrence of acute ischemic stroke,while in females the relationship reverses.Both correlation is independently from high blood pressure,coronary heart diseases,BMI,homocysteine,total cholesterol,total triglyceride,low density lipoprotein,media thickness of carotid artery,as well as the existence of plaques in carotid artery.2.Independently from risk factors mentioned above,the missense mutation of rs2274907(T>A)might bring down the acute ischemic stroke occurrence irrespective of gender difference. |
