| Objective:To observe the effect of Yiqi Huoxue Decoction on influences and quality of life of patients with lung cancer after radiotherapy,bone marrow function,immune function and tumor immune microenvironment,to investigate the effects of Yiqi Huoxue Prescription on the body of patients with lung cancer radiotherapy for body immune and tumor immune microenvironment.Methods:Collected from september 2015 to December 2016 in China people’s Liberation Army 251 st hospital for the first time of radiotherapy undergoing three-dimensional conformal radiotherapy(3D-CRT)or intensity-modulated radiotherapy(IMRT),and 120 cases of lung cancer were confirmed by pathology.According to the random number table,the patients were randomly divided into the control group(only radiotherapy)60 cases,the treatment group(traditional Chinese medicine plus radiotherapy)60 cases,and finally eliminated the fall off statistics,selected 112 cases: 56 cases in the control group and 56 cases in the treatment group.There were no significant differences between the two groups in general datas.According to the recent curative effect,QLQ-LC13,bone marrow function,transforming growth factor-beta 1,Interleukin-10,CD4(+)CD25(+)regulatory T lymphocytes and CD4(+)CD25(+)Foxp3(+)Tregs as indexes to observe the effect of Yiqi Huoxue Decoction before and after radiotherapy and radiotherapy for lung cancer patients with radiotherapy for body immune and tumor immune microenvironment.SPSS 21 statistic software was used to analyse the outcomes for experimental datasResults:1 The concentration of TGF-β1and IL-10 in blood serum(1)The concentration of TGF-β1from the control group and treatment group before radiotherapy and radiotherapy 20 Gy,P values were 1.000 and 0.414,there was no significant difference(P>0.05),except the time points P value were 0.000,the differences between groups were statistically significant(P<0.05).the average concentration of TGF-β1 is the lowest(568.09ng/L),when the treatment group was treated with 2 months after radiotherapy.The lowest in the control group before radiotherapy(589.893ng/L).Repeating the measurement time and the interaction levels between groups and individual effects: F-measure was 35.145,P value was 0.000,time of repeated measurements and interaction between groups.The measuring point average highest concentration TGF-β1of treatment group and control group were at the end of radiation(597.214ng/L and 700.430ng/L).(2)The concentration of IL-10 between the treatment group and control group before radiotherapy and radiotherapy 20 Gy,P values were 0.884 and 0.751,there was no significant difference(P>0.05),more than the time points P values were 0.018,0.000,0.002 and 0.000.the differences between groups were statistically significant(P<0.05),2 month after radiotherapy the average concentration of IL-10 of the treatment group to the lowest(22.427ng/L),the control group was the lowest before radiotherapy(25.406ng/L).Repeating the measurement time and interaction levels in the two groups and individual effects: F-measure was 83.382,P value was 0.000,the time of repeating measurements and the interactions between the two groups,the treatment group and control group at the end of the measuring point in radiotherapy,the average concentration of IL-10 was the highest(27.714ng/L and 35.610ng/L).2 Expression of CD4(+)Tregs in peripheral blood(1)CD4(+)CD25(+)Tregs content between the control group and treatment group before radiotherapy,P value was 0.623,the difference was not statistically significant(P>0.05).more than the time points,P values were 0.000,0.000,0.000,0.477 and 0.438,groups were statistically significant(P<0.05),CD4(+)CD25(+)Tregs was the lowest in the treatment group 2 month after radiotherapy(19.513%),the control group was the lowest before radiotherapy(20.015%).Repeating the measurement time and interaction levels between the two groups and individual effects: F-measure was 883.191,P value was 0.000,the time of repeating measurements and groups interaction,when the measurement point after radiotherapy,the content of CD4(+)CD25(+)Tregs were the highest between the treatment group and control group(22.507% and 32.988%).(2)CD4(+)CD25(+)Foxp3(+)Tregs was the lowest between the treatment group and control group before radiotherapy(5.707% and 5.810%),the P value was 0.694,there was no significant difference(P>0.05),except the time points,P values were 0.005,0.009,0.000,0.000 and 0.000,the differences between groups were statistical significance(P<0.05).Repeating the measurement times and interaction levels between the two groups and individual effects: F-measure was 67.776,P value was 0.000,indicating the existence of interaction between groups and repeating measurement times.measurement points was the highest,when the treatment group was treated during 40 Gy radiotherapy(6.432%),the content of measuring point CD4(+)CD25(+)Foxp3(+)Tregs was the highest in the control group at the end of radiotherapy(8.221%).3 The degree of bone marrow suppression of the two group,at a dose of 40 Gy during radiotherapy,the end of radiotherapy and both 1month and 2 month after radiotherapy,P values were 0.017,0.000,0.000,0.000,there was significant difference(P<0.05);before radiotherapy and at 20 Gy radiotherapy,P values were 0.799 and 0.295,there was no significant difference(P>0.05).Compared to the treatment group before radiotherapy: both 1month and 2 month after radiotherapy,P were 0.014 and 0.002,there was statistically significant(P<0.05);the control group was at 40 Gy radiotherapy,the end of radiotherapy January,and both 1month and 2 month after radiotherapy,P values were 0.008,0.000,0.000,0.000,the difference had statistical significance(P<0.05)4 The short-term curative effect of the two groups was better than before treatment.The treatment group CR14 cases,PR27 cases,SD12 cases,PD3 cases,ORR was 73.70%,DCR was 94.60%;the control group CR5 cases,PR18 cases,SD22 cases,PD11 cases,ORR 41.10%,DCR 80.40%.Compared with the control group,the ORR and DCR of the treatment group under the intervention of Yiqihuoxue decoction were significantly better than the control group,the P values were 0.001 and 0.022 respectively,and the difference was statistically significant(P<0.05).5 The QLQ-LC13 scores of the two groups increased to some extent from the beginning of radiotherapy to the end of radiotherapy,and there was a significant difference at 20 Gy radiotherapy,F was 17.363,P value was 0.000.Repeating the measurement times and interaction levels in the two groups and individual effects: F-measure was 127.940,P value was 0.000,indicating the existence of interaction between groups and repeated measurement times,the average scores of measurement QLQ-LC13 reached the peak at the end of radiation,the treatment group and control group were 26.741 and 38.883,F was 321.432,P value was 0.000,with significant difference between the treatment group(P<0.05);the average score of the minimum was 19.889 at 2 month after radiotherapy,the lowest was 20.591 in the control group before radiotherapy.Conclusions:1 Yiqihuoxue recipe could reduce the immunosuppression caused by radiotherapy in tumor microenvironment with increasing the expression of IL-10,TGF-β1 and CD4(+)Tregs,and enhance the efficacy of radiotherapy.2 As Foxp3 was Tregs specific marker,Tregs was associated with the occurrence and development of lung cancer patients with radiotherapy.3 Yiqihuoxue decoction could reduce the incidence of bone marrow suppression,reduce the degree of bone marrow suppression,and play a role on reducing toxicity and increasing effectiveness of lung cancer patients with radiotherapy.4 Yiqihuoxue decoction combined with radiotherapy could effectively improve the quality of life and control the progress of lung cancer patients. |
