A novel multi-functional nanoparticle(DTX/PFH@PPy-FA)with tumor active targeting function and visualized photothermal-chemotherapy combined therapeutic effect was constructed.Lipid-perfluorocarbon nanonanospheres loaded with DTX was firstly fabricated by solvent evaporation-emulsion method,then Lipid-perfluorocarbon nanonanospheres were coated with PPy shell.Finally,the folic acid molecules with active tumor-targeting function were modified on the surface of PPy shell.The DTX/PFH@PPy-FA was uniform in size,with a zeta potential of 8.95 ± 1.08 mV,a particle size of 209.1 ± 7.2 nm,and a drug loading of 19.5± 0.8%.DTX/PFH@PPy-FA possessed good photothermal effect,photoacoustic imaging effects and ultrasound imaging effects in vitro.MTT assay indicated that the blank PFH@PPy-FA vehicle showed low cytotoxicity to 4T1 and Hela cells.MTT assay also indicated the DTX/PFH@PPy-FA had good active targeting ability to tumor cells and its photothermal and photothermalchemotherapy synergistic effects showed excelent inhibition to tumor cells.The BALB/c 4T1 breast cancer model was built,and the in vivo imaging function and anti-tumor effect of the DTX/PFH@PPy-FA were studied.The experimental results showed that DTX/PFH@PPy-FA had good ultrasound imaging and photoacoustic imaging function in vivo.Photothermalchemotherapy synergistic effect showed good antitumor effect in vivo and could significantly inhibit lung metastasis of 4T1 breast cancer.The multifunctional nanopasrticle is expected to be a new type of visualized photothermal therapeutic agent.
